A low-cost and reconfigurable autonomous microscopy platform is provided capable of automated slide scanning and correlated brightfield and fluorescence imaging. Method of spectral imaging using the platform with applications including infectious disease diagnosis is also provided herein.

Spectral imaging platform for infectious disease diagnosis

Instrument free 'plasticware' is provided that enables multi-step molecular reactions in a diagnostics context. A hollow plunger that is movable within a surrounding tube defines a reaction chamber inside the plunger. By moving the plunger to different positions in the tube, a sample can be collected, then the sample can be washed, and finally the reaction chamber can be sealed to perform diagnostic reactions on the sample. The juxtaposition of large sample collection/washing volume with small reaction volume allows one to conduct a wide range of diagnostic assays including a LAMP (Loop mediated isothermal amplification) based saliva test in a small, portable self-contained device. Applications include Molecular diagnostics for health applications (including COVID19 test), Environmental Monitoring, Disease surveillance, and Veterinary health.

A microfluidic centrifuge is provided that includes a microfluidic disc having a pair of through holes that include a first through hole and a second through hole, where the pair of through holes are disposed symmetric and proximal to a central axis of the microfluidics disc, where the microfluidics disc includes a sealable input port and a sealable output port, where the sealable input port and the sealable output port are connected by a microfluidics channel, and a pair of tethers disposed through the pair of through holes that are disposed to twist about each other for unwinding where the tether unwinding is disposed to interact with the through holes, where the microfluidics disc rotates about the central axis.

Paperfuge: An integrated paper-based centrifugation and microfluidics platform for low-cost diagnostics

An annular fluid-filled sample chamber is used to provide observation of unbounded motion of objects in the fluid. Rotation of the sample chamber is under automatic control to compensate for azimuthal motion of the object, thereby keeping the object in a fixed field of view of an optical observation system. Further motion control can be provided in the radial and focus directions, which can be used to provide full 3D tracking of objects as they move in the fluid. An important application of this work is to observation and tracking of objects that move up or down in the fluid with respect to gravity.

Hydrodynamic treadmill: a tracking device to study biotic/abiotic systems in gravitational and hydrodynamic fields

A magnetic trap droplet controller is provided that includes a solid substrate, a controller operated by an appropriately programmed computer, a two-dimensional distribution of magnetized domains disposed on a surface of the solid substrate, a fluid chamber disposed above the two-dimensional distribution of magnetized domains, and an active magnet, where the active magnet is disposed to provide a dynamic magnetic field in-plane with the solid substrate, where the dynamic magnetic field controls north and south poles of the two-dimensional distribution of magnetized domains according to the controller, where a fluid under test that is disposed in a carrier fluid that is disposed in the fluid chamber propagates according to the controlled north and south poles of the two-dimensional distribution of the magnetized domains.

Two-dimensional magnetic trap arrays for droplet control

An optical device, such as a microscope, is disclosed that can be assembled from flat materials. The optical device can be assembled via a series of folds of a flat material. The optical microscope can include a stage for supporting a sample, an optic stage, and a light source. The optic stage can include one or more lenses. The optical microscope can be capable of obtaining simultaneous images from different forms of microscopy. The optical microscope may have bright field and filter field viewing capabilities wherein a user shifts from bright field to filter field by lateral movement of the stage containing a lens and a light source that cooperate to provide either the bright field or the filter field.

Optical microscope

Techniques are provided for a completely programmable fluidic manipulation without requiring any external control elements or electricity. A chip includes multiple microfluidic channels separated from an outside of the chip by an outer flexible substrate. An apparatus includes a plurality of actuators rotatably connected to a support structure with a recess for receiving the chip. Each actuator includes a plurality of teeth protruding outward, and is positioned so that at least in some angle of rotation a tooth of the actuator extends sufficiently into the recess to compress a microfluidic channel in a chip disposed in the recess. An optional punch card guide is included for guiding a card to contact the actuators. A system or kit includes a punch card on which is formed a plurality of punch features, each configured to engage a tooth of an actuator. Different microfluidic protocols are executed by simply changing the punch card.

A magnetic-hydrodynamic force fluid logic controller is provide that includes a solid or flexible or flexible substrate, a fluid chamber disposed above the substrate, where the chamber includes a fluid under test that includes an active magnet, where the active magnet is disposed to control a magnetic north pole of the droplet and a magnetic south pole of the droplet, a two-dimensional distribution of magnetic elements a surface the solid or flexible substrate, where the magnetic elements comprise a magnetization in a magnetic north pole and magnetization in a magnetic south pole, where the magnetic elements are activated by an external magnetic field of the active magnet, where the droplets have a droplet magnetization, where the droplet magnetization is configured for droplet self-interaction by the magnetic elements and the active magnet, where the self-interaction comprises splitting, merging, propagation, logic, storage, memory and all possible combinations of logical circuit operations.

Synchronous universal droplet logic

A permanently sealed vacuum tube is used to provide the electrons for an electron microscope. This advantageously allows use of low vacuum at the sample, which greatly simplifies the overall design of the system. There are two main variations. In the first variation, imaging is provided by mechanically scanning the sample. In the second variation, imaging is provided by point projection. In both cases, the electron beam is fixed and does not need to be scanned during operation of the microscope. This also greatly simplifies the overall system.

Optical lenses and methods for manufacturing optical lenses are disclosed herein. Lenses having a reduced aperture size are also disclosed herein along with methods for making the same. The optical lenses disclosed herein can be made having improved optical properties. The lenses can be used in optical microscopes, including optical microscopes with a shorter optical path relative to conventional optical microscopes.

Optical lens fabrication

A method, system and apparatus for imaging an oral cavity of a subject include a bracket comprising a mouthpiece and a camera mount. The mouthpiece has upper and lower bite guides disposed on a posterior side and separated by an opening through the mouthpiece. The bite guides are spaced apart so a subject biting on the guides opens the oral cavity to inspection through the opening. The mount is disposed on an anterior side of the mouthpiece; and has a flange to engage and slide along the opening and an optical path for light to pass through the mount and mouthpiece. A clip on an anterior side of the mount is configured to removeably hold a camera to record light passing through the optical path from the posterior side of the mount. In one embodiment, the camera is a programmable cell phone with digital camera.
