A gene construct comprising an FbLate promoter is disclosed. A method of obtaining an FbLate promoter and a plant, cell and seed comprising an FbLate promoter are also disclosed.

FbLate promoter

An isolated cotton fiber-specific promoter is disclosed. Preferably, this promoter comprises a nucleotide segment comprising at least 2.7 kb of the upstream genomic of a gene of the E6 family, or 1.7 kb of the upstream genomic family of the FbLate family.

Cotton fiber-specific promoters

A method is disclosed which describes the identification of cDNA clones useful for identifying fiber genes in cotton. The cDNA clones are useful in developing corresponding genomic clones from fiber producing plants to enable genetic engineering of cotton and other plants using these genes. The fiber-specific genes are identified by differential cDNA library screenings.

Genetically engineering cotton plants for altered fiber

A fiber-producing plant comprising in its genome a heterologous genetic construct is disclosed. This genetic construct comprises a fiber-specific promoter and a coding sequence encoding a plant peroxidase. Preferably, the coding sequence is for cotton peroxidase. Seeds of the plant containing this genetic construct and plant cells containing this construct are also disclosed.

Transgenic cotton plants producing heterologous peroxidase

A method of genetically engineering a fiber-producing plant is disclosed. This method first comprises the step of constructing a plant expression vector that comprises a protein coding sequence and a DNA sequence capable of promoting gene expression in fiber cells, wherein the DNA sequence is homologous to a sequence selected from the group consisting of 12 different genomic sequences. The method next involves introducing the expression vector into a fiber-producing plant wherein the protein coding sequence is expressed in the fiber cells of the fiber producing plant. A method of obtaining a DNA sequence capable of promoting gene expression in fiber cells is also disclosed.

A fiber-producing plant comprising in its genome a heterologous genetic construct is disclosed. This genetic construct comprises a fiber-specific promoter and a coding sequence selected from the group consisting of sequences encoding bioplastic-producing genes. Preferably, the coding sequence is selected from the group consisting of ketothiolase, acetoacetyl-CoA reductase, and PHB synthase. Seeds of the plant containing this genetic construct and plant cells containing this construct are also disclosed.

A method is disclosed which describes the identification of cDNA clones useful for identifying fiber genes in cotton. The cDNA clones are useful in developing corresponding genomic clones from fiber producing plants to enable genetic engineering of cotton and other plants using these genes. The fiber-specific genes are identified by differential cDNA library screenings. Coding sequences from these isolated genes are used in sense or antisense orientation to alter the fiber characteristics of transgenic fiber-producing plants.
