A microfluidic chip for causing the delivery of a payload to a cell comprises a plurality of constrictions configured to allow a cell suspension to flow through one or more of the plurality of constrictions from a first fluid flow region to a second fluid flow region within the microfluidic chip, wherein a cross-sectional width of each of the plurality of constrictions is less than a diameter of cells in the cell suspension, such that membranes of the cells are perturbed when passing through the constrictions such that a payload is able to pass through the perturbed cell membranes, and wherein a quotient of a cross-sectional area over a cross-sectional perimeter of each of the plurality of constrictions is greater than greater than or equal to 0.5 μm.

High-throughput microfluidic chip having parallelized constrictions for perturbing cell membranes

A system for delivering a payload to a cell that includes: a platform supporting an input container, an output container, and a receiver for receiving all or part of a disposable assembly, the disposable assembly including a preparation vessel and a constriction cartridge. The preparation vessel holds a cell suspension as it is prepared for passage through one or more cell-deforming constrictions, and the constriction cartridge houses a component that includes the one or more cell deforming constrictions. Passage through the cell-deforming constrictions causes perturbations in cell membranes to allow entry of a payload into the cells. The system includes one or more processors configured to receive input from a user and to automatically control pressure, temperature, agitation, and/or flow of the cell suspension as it passes through the input container, through the preparation vessel, through the constriction cartridge, and to the output container.

System for delivery of a payload into a cell

The present application provides formulations of modified peripheral blood mononuclear cells (PBMCs), wherein the formulation comprises: PBMCs comprising at least one antigen (e.g., a human papillomavirus (HPV) antigen) and a cryopreservation medium. In some embodiments, the PBMCs are conditioned by incubating the PBMC in the presence of an adjuvant.

A cartridge for delivering a payload to cells of a cell suspension is provided, wherein the cartridge comprises an input channel that delivers the cell suspension to a first plurality of branch channels, and wherein the first plurality of branch channels each deliver the cell suspension into a respective one or a plurality of microfluidic chips or filters. Cell suspension exiting a microfluidic chip or filter flows into a respective one of a second plurality of branch channels, and is then delivered to an output channel by which it exits the cartridge. The cartridge may comprise a plurality of removable covers that hold the chips or filters in place against a body of the cartridge in which the input channel, output channel, and branch channels are formed.

Cartridge for use in a system for delivery of a payload into a cell

The present invention generally relates to receiving bodily fluid through a device opening. In one aspect, the device includes a flow activator arranged to cause fluid to be released from a subject. A deployment actuator may actuate the flow activator in a deployment direction, which may in turn cause fluid release from a subject. The flow activator may also be moved in a retraction direction by a retraction actuator. In one aspect, the device may include a vacuum source that may help facilitate fluid flow into the opening of the device and/or may help facilitate fluid flow from the opening to a storage chamber. In one aspect, a device actuator may enable fluid communication between the opening and the vacuum source and the flow activator may be actuated after the enablement of fluid communication.
