The present invention relates to novel subtilase variants exhibiting alterations relative to the parent subtilase in one or more properties including: Wash performance, thermal stability, storage stability or catalytic activity. The variants of the invention are suitable for use in e.g. cleaning or detergent compositions, such as laundry detergent compositions and dish wash compositions, including automatic dish wash compositions.

Subtilase variants

The present invention relates to methods for producing a polypeptide having biological activity, comprising: (a) cultivating a fungal host cell in a medium conducive for the production of the polypeptide, wherein the fungal host cell comprises a first polynucleotide encoding the polypeptide operably linked to a second polynucleotide encoding a variant signal peptide or a variant prepropeptide; and (b) isolating the secreted polypeptide having biological activity from the cultivation medium.

Methods for producing secreted polypeptides having biological activity

The present invention relates to pectate lyase variants exhibiting alterations relative to a parent enzyme exhibiting pectate lyase activity as its major enzymatic activity; to a method of producing such enzymes; and to methods for using such enzymes in the textile, detergent and cellulose fiber processing industries. Compared to the parent enzyme, the pectate lyase variants of the present invention exhibit improved stability in detergents.

Pectate lyase variants

The present invention relates to methods for producing a polypeptide, comprising: (a) cultivating a fungal host cell in a medium conducive for the production of the polypeptide, wherein the fungal host cell comprises a first polynucleotide comprising a nucleic acid sequence encoding the polypeptide operably linked to a copper-inducible promoter sequence comprising a copper-responsive upstream activation sequence activated by a copper-dependent trans-acting transcription factor and a second polynucleotide comprising one or more (several) additional copper-responsive upstream activation sequences operably linked upstream to the promoter sequence, wherein the promoter sequence is foreign to the nucleic acid sequence encoding the polypeptide and the copper-responsive upstream activation sequences are responsible for copper-induced transcription of the promoter sequence, and a third polynucleotide comprising at least one copy of a gene encoding the copper-dependent trans-acting transcription factor; and (b) isolating the polypeptide from the cultivation medium.

The invention provides variant lipases, preferably, variants with reduced tendency to odor generation obtained by introducing mutations in one or more regions identified in a parent lipase.

Lipase variants

The present invention relates to mannanases comprising e.g., a sequence of amino acids 32&#x2013;330 of SEQ ID NO: 2 or their homologues may be derived from e.g.,sp. I633, or may be encoded by polynucleotide molecules comprising a sequence of nucleotides 94&#x2013;990 of SEQ ID NO: 1 from, polynucleotide molecules that encode a polypeptide that is at least 65% identical to the sequence of amino acids 32&#x2013;330 of SEQ ID NO: 2, or degenerate nucleotide sequences thereof. The mannanases are alkaline and are useful e.g. in cleaning compositions, in a fracturing fluid useful to fracture a subterranean formation, for modifying plant material, and for treatment of cellulosic fibers.

Mannanases

The present invention relates to pectate lyases comprising the amino acid sequence Asn Leu Asn Ser Arg Val Pro (NLNSRVP) (amino acids 236&#x2013;242 of SEQ ID NO: 2) belonging to Familyof polysaccharide lyases have good performance in industrial processes under neutral or alkaline conditions such as laundering and textile processing. The pectate lyase may be derivable fromspecies.

Pectate lyases

The present invention relates to an enzyme exhibiting endo-beta-1,4-glucanase activity (EC 3.2.1.4), which is a) a polypeptide encoded by the DNA sequence of positions 1 to 2322 of SEQ ID NO: 1; b) a polypeptide produced by culturing a cell comprising the sequence of SEQ ID NO: 1 under conditions wherein the DNA sequence is expressed; c) an endo-beta-1,4-glucanase enzyme having a sequence of at least 97% identity to the amino acid sequence of position 1 to position 773 of SEQ ID NO: 2; and fragments thereof exhibiting endo-beta-1,4-glucanase activity, and d) a polypeptide having endo-beta-1,4-glucanase activity that is encoded by a polynucleotide that hybridizes with the nucleotide sequence shown in positions 1&#x2013;2322 of SEQ ID NO: 1, is useful for detergent and textile applications.

Endo-beta-1,4-glucanases

The invention relates to a method of identifying enzymes suitable for use in detergents, especially the selection of specific enzyme variants among a large number of such variants created through random mutagenesis. The method comprises that enzyme variants to be selected are in a mixture of enzyme variants which are each displayed on the surface of cells or phage particles, and (i) introducing the mixture into a detergent composition under conditions that will have a negative impact on the activity of or inactivate most of said enzyme variants, (ii) reacting said mixture with a catcher molecule that will bind specifically only to enzyme variants that exhibit the property sought for, (iii) separating said complex from the remaining parts of said mixture, (iv) dissociate said complex to isolate such cell(s) or phage(s) that displayed said enzyme variant without the catcher molecule, (v) introducing said phage into a host wherein it will multiply, or (va)cultivating said cell under conditions conducive to its multiplication, (vi) isolating a DNA molecule coding for said enzyme variant from the genome of said cell or phage, and (vii) determining the sequence of said DNA molecule. The invention further relates to specific materials used in said method, the enzymes selected by use of the method, and to detergent compositions comprising aid enzymes.
