Transdermal drug delivery patches and methods of their production are described. The patches can be made such that the accommodate highly plasticizing drugs such as selegiline and/or the use of protonated forms of various drugs.

Adhesive mixture for transdermal delivery of highly plasticizing drugs

A method for delivery of substance through at least one dermal layer, by providing a substance in microcapsules at a predetermined size, within a medium () for holding the microcapsules; placing the medium for holding the microcapsules on a surface of a patch () adjacent the skin () of a human or animal; and applying energy () to the patch, the energy having a characteristic of disturbing the integrity of the microcapsules, thereby resulting in release of the substance from the microcapsules. The energy may be selectively applied to release the substance at desired times. The substance may be a drug or other active agent.

Transdermal delivery using encapsulated agent released by ultrasound and/or heat

An apparatus for sonoporation for transdermal delivery of a microparticles suspension containing microencapsulated drugs includes a container containing said microparticles suspension and an ultrasound horn having a tip submerged in said microparticles suspension containing microencapsulated drug or the like. The ultrasound radiation is applied to generate cavitation bubbles, thus causing pores to be formed in the skin of a patient. The ultrasound radiation intensity and distance from the skin are also effective in generating ultrasonic jets driving the microparticles through the formed pores into the skin. The ultrasound radiation is desirably applied at a frequency other than a resonant frequency of the microparticles to avoid rupturing them.

Intradermal incorporation of microparticles containing encapsulated drugs using low frequency ultrasound

An apparatus for performing in-vivo sonoporation of a skin area and transdermal and/or intradermal delivery of a drug solution includes a container having an end adjacent the skin area and containing the drug solution. The container further includes an ultrasound horn having a tip submerged in the drug solution for applying ultrasound radiation to the drug solution. The ultrasound radiation has a frequency in the range of 15 KHz and 1 MHz and is applied at an intensity, for a period of time and at a distance from said skin area effective to generate cavitation bubbles. The cavitation bubbles collapse and transfer their energy into the skin area, thus causing the formation of pores in the skin area. The ultrasound radiation intensity and distance from the skin area are also effective in generating ultrasonic jets, which ultrasonic jets then drive the drug solution through the end adjacent the skin area and the formed pores into the skin.

Method and apparatus for in-vivo transdermal and/or intradermal delivery of drugs by sonoporation

An apparatus for sonoporation for transdermal delivery of a microparticles suspension containing microencapsulated drugs includes a container having an end covered with a porous membrane and containing said microparticles suspension and an ultrasound horn having a tip submerged in said microparticles suspension containing microencapsulated drugs. The ultrasound horn applies ultrasound radiation to the microparticles suspension containing microencapsulated drugs. The ultrasound radiation is applied at a frequency, an intensity, for a period of time, and at a distance from the skin, effective to generate cavitation bubbles, wherein the cavitation bubbles collapse and transfer their energy into the skin area thus causing the formation of pores in the skin area; and wherein the ultrasound radiation intensity and distance from the skin area are also effective in generating ultrasonic jets, the ultrasonic jets driving the microparticles suspension containing microencapsulated drugs through a porous membrane and the formed pores into the skin area. The ultrasound radiation can optionally have a frequency of 1 KHz to 5 MHz.

Method and apparatus for intradermal incorporation of microparticles containing encapsulated drugs using low frequency ultrasound

Devices for the controlled release of an active agent to the skin or mucosa of a host are disclosed, which devices are laminates of a backing layer and a monolithic carrier layer formed from a melt blend of an active ingredient with a thermoplastic matrix polymer without first dissolving or suspending the polymer or active ingredient in a solvent therefor, so that the carrier layer is substantially free of residual solvent, wherein the thermoplastic matrix polymer has a melt temperature between about 170.degree. C. and about 200.degree. C. and is selected from polyether block amides, ethylene methacrylic acid copolymers, ethylene acrylic acid copolymers, copolymers of polyether prepolymers with polybutylene terephthalate, copolymers of polyether prepolymers with polyisobutylene terephthalate and polyether polyurethanes, the active ingredient is heat stable at the melt temperature of the matrix polymer and is selected from active agents, active agent enhancers and mixtures thereof, and the laminate is provided with means for affixing the laminate to the skin or mucosa of the host so that the active ingredient is continuously released from the carrier layer thereto. Devices for the controlled release of an active agent to the skin or mucosa of a host are disclosed, which devices are laminates of a backing layer, a monolithic carrier layer of an active ingredient selected from active agent, active agent enhancers and mixtures thereof, melt-blended with a thermoplastic matrix polymer capable of controllably releasing the active ingredient, wherein the active ingredient is heat stable at the melt temperature of the matrix polymer, together with means for affixing the laminate to the skin or mucosa of the host so that the active ingredient is continuously released from the carrier layer thereto. Methods for assembling the device are also disclosed, in which a thermoplastic matrix polymer capable of controllably releasing an active ingredient is melt-blended with an active ingredient that is heat stable at the melt temperature of the matrix polymer, so that a melt-blend of the active ingredient and the thermoplastic matrix polymer is formed, which melt-blend is formed into a monolithic carrier layer, and the carrier layer is then combined with a backing layer and means for affixing the laminate to the skin or mucosa of the host so that the active ingredient is released from the carrier layer thereto.

Devices for the controlled release of an active agent to the skin or mucosa of a host are disclosed, which devices are laminates of a cellular foam layer having a first surface and a second surface, which foam layer has an active agent incorporated therein; a backing layer having an inner surface and an outer surface, wherein the inner surface is affixed to the second surface of the foam layer so that the active agent cannot permeate from the second surface of the foam layer through the outer surface of the backing layer; and means for affixing the laminate to the skin or mucosa of the host so that the active agent is capable of being continuously released from the first surface of the foam layer thereto. Methods for assembling the laminate devices are also disclosed, as are methods in which the foam layer is formed from a prepolymer solution on the backing layer. Methods are also disclosed for the polymerization of cellular urethane foams having active agents uniformly dispersed therethrough.
