Methods, devices, and systems are disclosed for performing high throughput analysis of conformational change in biological molecules or other biological entities using surface-selective nonlinear optical detection techniques.

Systems and methods for high throughput analysis of conformation in biological entities

Instrument and method for detecting pesticides and other analytes. A sample to be tested is mounted on a cassette and inserted into a housing which is substantially impervious to light, and light from a source within the housing is directed toward the sample to induce fluorescent emission from analyte on the sample. Fluorescent emissions from the sample are monitored with a detector within the housing to detect emissions having a spectral content characteristic of the analyte to be detected. Data from the detector is processed, and information based on the processed data is displayed. In some embodiments, the instrument is calibrated with data from a reference sample of known concentration or density. The detector measures the analyte in units of mass per unit area, and in some embodiments the mass per unit area is converted to units of concentration or density of the analyte in the sample.

Analyte detector and method

The present invention provides an improved integrated system for biological marker identification. The system uses Microvolume Laser Scanning Microscopy (MLSC) in order to measure patterns of expression of biological markers in biological fluids. The system includes improved instrumentation for performing MLSC, and also includes improved particle detection and analysis methods. The system further comprises an informatics architecture for the analysis of data obtained from the MLSC in tandem with other medical information.

A disposable cuvette cartridge for optical measurements is formed from three flexible thin layers. The middle layer is an adhesive sheet with cut out regions, typically between 30 and 50, serving as optical chambers bounded by the two outer layers. Inlet and vent holes in one of the outer sheets provide access to the optical chambers. At least one of the outer sheets is made of a material having low fluorescence, such as a cyclo-olefin copolymer. With its low background fluorescence, the cuvette cartridge is useful for high-throughput fluorescence measurements of biological samples.

A cuvette cartridge for optical measurements is disclosed herein, the cartridge comprising a flat, adhesive sheet having a selected thickness that defines an optical path length perpendicular to the longitudinal axis of such sheet; wherein a portion of the adhesive sheet is cut out to define at least one optical chamber of a desired length; wherein the adhesive sheet is placed between a first flat sheet and a second flat sheet, the first flat sheet comprising at least one inlet hole and at least one outlet hole, the vent hole being located at a first end of the optical chamber and the inlet hole being located at a second end of the optical chamber. An integrated analytical system employing this cartridge and optionally including a vacuum chuck for holding and registering the cartridge are also disclosed.

A method and an apparatus for analyzing a material within a container, such as blood within a capillary in a volumetric cytometry system provides for detecting the edges of the container, counting the cells within the container, characterizing the cells within the container, and evaluating channels of data which contain information relevant to more than one of the detectable characteristics of the cells. A scanner scans a container of material including certain cells. Sampling circuitry is coupled to the scanner to generate scanned images of the material in the container. Two or more scanned images are generated based on fluorescence data from dyes that have overlapping spectra. The two scanned images are processed using a linear regression analysis among corresponding pixels in the scanned images near certain cells to characterize relative contents of two fluorescing dyes in a target cell. Target cells are identified from the scanned images using processing resources which identify a peak sample within a neighborhood, and compare the amplitude of the peak with the amplitude of pixels on the perimeter of the neighborhood. Upon identifying a target cell in this manner, data from the plurality of scanned images corresponding to the identified cell are saved for further analysis.

Method and apparatus for cell counting and cell classification

A frequency synthesizer comprises a voltage controlled oscillator (VCO) with an output frequency f.sub.out that is dependent on a VCO-control voltage input. A divide-by-k counter is connected to receive the output frequency f.sub.out and to output a pair of in-phase and quadrature phase signals f.sub.k. A vector modulator is connected to receive the in-phase and quadrature phase signals f.sub.k and the output frequency f.sub.out, and outputs a single sideband (SSB) modulated output f.sub.n comprising only one of the sum (f.sub.out +f.sub.k) or difference (f.sub.out -f.sub.k) products. A divide-by-n counter is connected to receive the modulated output f.sub.n and to output a feedback frequency sample f.sub.f. And a phase detector is connected to receive the feedback frequency sample f.sub.f and to compare it with a reference frequency f.sub.r. A phase error output signal is provided for the VCO-control voltage input.
