This invention describes a method of using controlled fluidic forces to improve the performance of a biochemical binding assay where a target molecule is captured by specific molecular recognition onto a substrate surface with an affinity coating, and then labeled with a detectable micrometer-scale particle using a second specific molecular recognition reaction with the target. By using specific ranges of label sizes and laminar flow conditions, controlled fluidic forces can be applied to the label particles in order to selectively remove molecules bound to a surface according to their binding strength, and thereby increase the ratio of specifically bound labels to more weakly attached non-specifically bound labels. This method can be used with a wide variety of label types and associated detection methods, improving the sensitivity and selectivity of a broad range of binding assays.

Fluidic force discrimination

Provided is a method and apparatus for attaching a fluid cell to a planar substrate. The planar substrate may have on it sensors or devices for detecting components within the fluid, and/or be treated to selectively bind or react with components within the fluid. Substrates might include solid-state IC integrated circuit sensor microchips, glass slides, genomic and proteomic arrays, and or other suitable substrates that can make conformal contact with the fluid cell. The fluid cell can be mounted directly on top of the substrate to easily create a fluidic system in a wide variety of implementations. The assembly does not require modification of the substrate; all the fluidic connections are inherent in the apparatus. The present device can be made using low-cost materials and simple methods.

Method and apparatus for attaching a fluid cell to a planar substrate

Provided is a method for controllably activating a surface for stable amine-reactive chemistries. A surface containing nitride is exposed to a plasma having a reactive species containing hydrogen for a period of time sufficient to activate the substrate for amine-reactive chemistries. Amine-reactive chemical processes can then be applied to the activated surface to reliably and controllably bond molecules directly to said surface. The method is designed to create stable primary amines on the nitride substrate, so that any subsequent amine-reactive chemistry may proceed in a controlled manner that is directly proportional to the density of surface amines so created.

The present invention describes an apparatus for nanolithography and a process for thermally controlling the deposition of a solid organic 'ink' from the tip of an atomic force microscope to a substrate. The invention may be used to turn deposition of the ink to the substrate on or off by either raising its temperature above or lowing its temperature below the ink's melting temperature. This process may be useful as it allows ink deposition to be turned on and off and the deposition rate to change without the tip breaking contact with the substrate. The same tip can then be used for imaging purposes without fear of contamination. This invention can allow ink to be deposited in a vacuum enclosure, and can also allow for greater spatial resolution as the inks used have lower surface mobilities once cooled than those used in other nanolithography methods.
