The present disclosure relates to an engineered immune cell and use thereof. The present disclosure provides an engineered immune cell comprising a CAR or engineered TCR, which CAR or engineered TCR can comprise a first antigen binding domain and a second antigen binding domain. The engineered immune cells of the present disclosure, when administered into a subject, can inhibit the host immune cells such as T cells and/or NK cells and enhance the survival and persistence of the engineered immune cells in vivo, thereby exhibiting more effective tumor killing activity.

Compositions and methods for T cell engineering

The present disclosure provides a method and system for quantifying a degree of blending of virgin and aged asphalt in HRAM. The method includes the following steps: first, constructing a relational equation between the microscale modulus of recycled asphalt in a fully blended state and the content of the aged asphalt; measuring the microscale modulus of the recycled asphalt, the microscale modulus of the aged asphalt, the microscale modulus of the virgin asphalt, and the content of the aged asphalt in the HRAM in situ; inputting the dates above into the relational equation to obtain the microscale modulus of the recycled asphalt in the fully blended state; and based on the microscale modulus of the recycled asphalt measured in situ and the microscale modulus of the recycled asphalt in the fully blended state, obtaining the degree of blending of the virgin and aged asphalt in the HRAM.

Method and system for quantifying degree of blending of virgin and aged asphalt in hot recycled asphalt mixtures (HRAM)

A preparation method of apolysaccharide rich in β-glucan includes: (1) extracting thepolysaccharide to obtain a filtrate; (2) concentrating the filtrate to obtain a supernatant; (3) precipitating a crudepolysaccharide from the supernatant using anhydrous ethanol to obtain a first precipitate; and (4) purifying the first precipitate to obtain thepolysaccharide. Thepolysaccharide rich in the β-glucan has stable quality, and its yield, polysaccharide content and β-glucan content are significantly improved compared with water extraction and alcohol precipitation methods in the related art. In vitro pharmacological experiments prove that thepolysaccharide rich in the β-glucan has good anti-inflammatory activity and can be applied to preparation of drugs or health foods.

Provided are a universal chimeric antigen receptor (CAR) T cell and preparation method and application thereof. The binding of the cell HLA-A/HLA-B to the TCR is inhibited; the TCR gene expression is silenced; the invention may be used for allogeneic tumor treatment and does not cause GVHD and HVG reaction during allogeneic infusion, and thus improves the survival and antineoplastic effect of allogeneic CAR-T cells in the recipient.

Universal chimeric antigen receptor T-cell preparation technique

The present invention provides an engineered immune cell targeting BCMA and use thereof. In particular, the present invention provides a CAR specifically targeting BCMA, the CAR comprising an antigen-binding domain which is an S-derived scFv, an antibody heavy chain variable region as shown in SEQ ID NO: 9 and an antibody light chain variable region as shown in SEQ ID NO: 10. The present invention also provides a CAR-T cell comprising the CAR, a double CAR- and CAR-T cell comprising the S-derived scFv, and related use thereof. Compared to CAR-T cells constructed using other scFvs, the constructed CAR-T cell of the present invention has a better killing effect and tumor elimination capability.

In the present invention, asphalt and aggregates are preheated separately, where the aggregates include coarse aggregates and fine aggregates; the coarse aggregates, part of the fine aggregates and asphalt that are preheated are subjected to a first mixing to obtain a first mixture; the remaining fine aggregates are added to the first mixture for a second mixing to obtain a second mixture; and a mineral powder is added to the second mixture for a third mixing, and discharging is conducted to obtain a hot-mixed asphalt mixture.
