Embodiments of a novel system for delivering an expression vector encoding an antigen to a subject that allows for spatiotemporal control over stimulation of the subject's immune response to the antigen are provided. In some embodiments, the expression vector delivery system includes a polymer linked to an adjuvant in prodrug form that can form polymer nanoparticles and enter a cell (such as an immune cell) under physiological conditions. In some embodiments, the adjuvant is linked to the polymer by an enzyme degradable labile bond, the cleavage of which activates the adjuvant to stimulate an immune response.

Expression vector delivery system and use thereof for inducing an immune response

A modified adenovirus, in particular Enadenotucirev (EnAd), armed with at least two bispecific T cell engagers each comprising at least two binding domains, wherein at least one of the domains is specific for a surface antigen on an immune cell of interest, such as a T-cell of interest. Also provided are a composition, such as a pharmaceutical formulation comprising the virus, use of the virus and virus formulations for treatment, such as in the treatment of cancer. The disclosure also extends to processes for preparing the virus.

Adenovirus armed with bispecific T cell engager

The invention disclosed herein relates to a method of cavitation-induced delivery of a therapeutic or diagnostic agent to a human or animal subject. In particular the invention provides an agent for use in a method of diagnosis or treatment of a human or animal subject, the method comprising exposing the subject to ultrasound, wherein the agent comprises a therapeutic or diagnostic component which is covalently bound to a dense component, the dense component having a density greater than that of the therapeutic or diagnostic component, and wherein either the dense component is a cavitation initiator or the method comprises administering to the subject a further agent which is a cavitation initiator. Binding of the dense component enhances cavitation-induced transport of the therapeutic or diagnostic component.

Sonosensitive therapeutic or diagnostic agent

Modified viral particles wherein the viral particles, typically adenoviral particles, are modified by glycosylation and the use of the modified viral particles to deliver heterologous nucleic acid to cells. Also disclosed are pharmaceutical compositions comprising the same and method of treatment using the same.
