Described herein is a microfluidic assay device that mimics in vitro the in vivo biological environment, supporting endothelization, allowing for perfusive flow similar to in vivo blood flow conditions, and providing for realistic interactions between T-cells and solid tumor cells, such as glioblastoma multiforme tumor cells. Also described herein are methods of using this microfluidic assay device for the study of interactions of immune cells with tumor cells, such as glioblastoma multiforme tumor cells, and the development of improved immunotherapeutic approaches against cancers, such as glioblastoma multiforme.

The present disclosure provides devices, kits, and methods for focusing/enriching and/or separating/sorting submicron size particles, including biological entities such as exosomes and other submicron size extracellular vesicles. Devices, kits, and methods for label-free focusing and/or separation of sub-micron particles

A variety of devices and methods are provided for separating or enriching circulating tumor cells in a biological sample such as whole blood. In some aspects, the devices are multi-stage devices including at least (i) a filtering stage, (ii) a sheath flow stage for ferrohydrodynamic separation of magnetically labelled white blood cells, and (iii) a focusing stage for marker-independent and size-independent focusing of magnetically labeled particles so as to separate or enrich unlabeled rare cells in the biological sample. The devices and methods are, in some aspects, capable of high throughput in excess of 6 milliliters per hour while achieving high separation (&#x3e;95%) of the unlabeled rare cells.

Devices and methods for separating circulating tumor cells from biological samples

A variety of devices, systems, kits, and methods are provided for separating or enriching circulating tumor cells in a biological sample such as whole blood. In general, embodiments of the present disclosure include non-uniform magnetic field-assisted processes and devices for the separation of particles (e.g., cells) within a magnetic fluid. Under non-uniform magnetic fields, particles such as cells can experience the generated magnetic field direction to produce a magnetic buoyancy force, analogous to buoyancy force, as magnitude of the force is proportional to the volume of cell. This force can be used to spatially separate cells of different sizes. In some embodiments, devices for separating particles are provided having a magnetic device configured to direct a non-uniform magnetic force onto the magnetic fluid and the particles; and a plurality of outlets, wherein the non-uniform magnetic force causes the types of particles to be separated and flow into different outlets.
