Compositions and methods for obtaining plant cells with modified Ht1 nucleotide sequences, modified NLB18 sequences, or both, are provided herein. The methods involve introducing double-strand breaks into the maize genome in an endogenous Ht1 encoding sequence, an endogenous NLB18 encoding sequence, or both, to modify the genomic sequence in order to enhance northern leaf blight resistance of a plant produced from the plant cell. Further provided are methods that introduce resistant alleles of Ht1 and/or NLB18 into specific sites in the genome. Plants produced by the plant cells, and seeds produced from the plants are also included. Guide polynucleotides are also provided for the use of the CRISPR-Cas system in inducing double strand breaks.

Compositions and methods are provided for the use of pollen-inhibitor genes and/or color marker genes in accelerated trait introgression. Compositions and methods are also provided for introducing a pollen-inhibitor gene and/or a color marker gene in close proximity to a trait locus of interest. Breeding methods and methods for selecting plants comprising a trait locus of interest in close proximity to at least one pollen-inhibitor gene and/or color marker gene are also disclosed. The methods and compositions employ at least one pollen-inhibitor gene and/or color marker gene to provide an effective system for accelerated trait introgression in the genome of a plant.

Methods and compositions for accelerated trait introgression

Compositions and methods useful in identifying and selecting maize plants with increased resistance to gray leaf spot are provided herein. The methods use molecular genetic markers within a QTL region located on chromosome 4 to identify and select plants with increased resistance to gray leaf spot, and plants comprising the QTL allele associated with increased resistance to gray leaf spot can be crossed to other maize plants to incorporate the increased resistance into other maize lines or varieties.
