Disclosed herein are methods in which an individual with multiple sclerosis (MS) can be classified into one of six subject groups, each subject group predictive for the patient's responsiveness to an interferon-&#x3b2; (IFN-&#x3b2;) therapy. The individual with MS can be classified according to the individual's serum marker levels, e.g., at baseline or following treatment with therapy. Depending on the classification, the individual with MS can be treated with standard therapies (e.g. IFN-&#x3b2;) or one or more alternative therapies with or without IFN-&#x3b2;.

Biomarkers for assessing subjects with multiple sclerosis

The instant disclosure provides replacement gene tolerizing vectors and methods of using such vectors for treating individuals receiving gene therapy for muscular dystrophy, e.g., Duchenne muscular dystrophy (DMD), Becker muscular dystrophy (BMD), etc. Aspects of the methods include administering to the individual, in need thereof, an effective amount of a gene therapy tolerizing vaccine composition that includes a replacement gene tolerizing vector to reduce one or more symptoms of a subject's immune response to gene therapy treatment for muscular dystrophy and/or improve the efficacy of the gene therapy. Compositions and kits for practicing the methods of the disclosure are also provided.

Replacement gene tolerizing vectors and methods of use thereof

The instant disclosure provides aquaporin DNA tolerizing vaccines and methods of using such vaccines for treating individuals having neuromyelitis optica (NMO) and NMO spectrum disorders. Aspects of the methods include administering to the individual, in need thereof, an effective amount of an aquaporin DNA tolerizing vaccine to reduce one or more symptoms of NMO or an NMO spectrum disorder. A pro-inflammatory T cell response is specifically suppressed by the injection into a recipient of DNA encoding an autoantigen associated with autoimmune disease. The recipient may be further treating by co-vaccination with a DNA encoding a Th2 cytokine, particularly encoding IL4. In response to the vaccination, the proliferation of autoantigen-reactive T cells and the secretion of Th1 cytokines, including IL-2, IFN-&#x3b3; and IL-15, are reduced.
