Provided are compositions for inhibiting a biological activity of an aldo-keto reductase family 1, member C3 (AKR1C3) polypeptide. In some embodiments, the compositions are indomethacin derivatives that are AKR1C3-specific inhibitors. Also provided are methods for producing disclosed indomethacin derivatives that substantially lack cyclooxygenase inhibitory activity but that have AKR1C3 inhibitory activity, methods for inhibiting AKR1C3 polypeptide biological activities, and methods for treating prostate tumors in subjects.

Indomethacin analogs for the treatment of castrate-resistant prostate cancer

The invention includes 2-&#x3b2;-naphthyl-acetic acid derivatives, which are selective AKR1C3 inhibitors. In certain embodiments, the compounds of the invention are (R)-naproxen analogs. The invention further includes methods of treating cancer, such as prostate cancer and/or castration-resistant prostate cancer, using at least one compound of the invention.

2-beta-naphthyl-acetic acid analogs as AKR1C3 inhibitors and methods of using same

A compound, a method of imaging a cell, and a method of detecting a cancer, an inflammatory disease, or a neoplastic disease are provided. The compound includes a nitroxide derivative of a fluorophore that is activatable upon exposure to free radicals. The method of imaging a cell includes contacting the cell with an activatable nitroxide derivative of a fluorophore. The method of detecting a cancer, an inflammatory disease, or a neoplastic disease includes administering an activatable nitroxide derivative of a fluorophore and then imaging one or more cells contacted with the compound.

Compounds and method for imaging cancer

A compound and method for detecting hypoxic cells and tissue are provided. The compound includes a probe selected from the group consisting of a hypoxia sensitive 2-nitroimidazole containing fluorescence imaging probe, a hypoxia sensitive reversible ON-OFF fluorescence imaging probe, a hypoxia sensitive azo-based fluorescence imaging probe, and combinations thereof. The method includes contacting the cells or tissue with the probe of any one of claims-and detecting fluorescent intensity of the cell or tissue, wherein increased fluorescent intensity indicates that the cells or tissue is hypoxic. Also provided are a method of synthesizing the compound and a method for synthesizing a therapeutic agent including the compound.

Composition and method for detecting hypoxia

Disclosed are compositions that comprise a cyclooxygenase-2-selective therapeutic and/or diagnostic agent having a therapeutic and/or diagnostic agent conjugated to a NSAID drug; and a ROS-responsive nanoparticle. Methods of making and using these compositions for drug encapsulation and delivery are also disclosed.

The presently disclosed subject matter provides compositions that selectively bind cyclooxygenase-2 and comprise a therapeutic and/or diagnostic moiety. Also provided are methods for using the disclosed compositions for diagnosing (i.e., by imaging) a target cell and/or treating a disorder associated with a cyclooxygenase-2 biological activity.

Methods and compositions for diagnostic and therapeutic targeting of COX-2

The presently disclosed subject matter provides derivatives of non-steroidal anti-inflammatory drugs (NSAIDs) that are characterized by substantially reduced cyclooxygenase inhibiting activity, yet retain the ability to interact with and modulate the activities of other polypeptides such as the class of peroxisome proliferators-activated receptors (PPARs) and &#x3b3;-secretase. Also provided are methods of using the derivatives to treat pathological disorders.

Indoleacetic acid and indenacetic acid derivatives as therapeutic agents with reduced gastrointestinal toxicity

The present invention provides methods, compositions and kits for discriminating between COX-1 and COX-2 activity. In particular, the prevent invention provides for the detection and/or measurement of COX-2 activity in subjects, samples thereof, and in laboratory tests. The present invention discloses that 2-arachidonylglycerol is a COX-2 selective substrate which is metabolized by COX-2 to prostaglandin glycerol esters (PG-Gs) and that the diversity of PG-Gs parallels that of arachidonic acid derived metabolites of COX. The present invention also provides certain novel COX-2 selective metabolites including prostaglandin I-glycerol ester (PGI-G) and 6-keto-prostaglandin F-glycerol ester. Methods and kits are described for detecting COX-2 activity comprising detecting PG-Gs (including the novel PG-Gs disclosed herein). Uses for these methods and kits include the detection and monitoring of inflammation and tumors or cancer. Additional uses include the monitoring of test agents in assays to screen for COX-2 specific inhibitors and other laboratory uses.

Compositions and methods for detecting and quantifying COX-2 activity and 2-arachidonylgycerol metabolites

A method of detecting an activity of a COX-2 enzyme in a subject that includes obtaining a sample of the subject; detecting an amino acid eicosanoid metabolite in the sample, wherein the presence of the amino acid eicosanoid metabolite indicates the activity of the COX-2 enzyme of the subject. Preferably the amino acid eicosanoid metabolite is a PGH-Gly or HETE-Gly metabolite. The metabolite may be detected based on metabolism of a COX-2-selective substrate. Preferably, the substrate is a lipoamino acid. More preferably, the lipoamino acid is selected from NAGly, N-arachidonyl-alanine, and &#x3b4;-arachidonyl aminobutuyic acid.
