The present invention relates to a process for purification of a target molecule, comprising the steps: (a) contacting a target molecule, and a population of target binding polypeptides (TBP), in solution for a sufficient time to allow complex formation; and (b) isolating the target from the complex from (a) by subsequent purification steps, wherein (i) the target binding polypeptides have at least two binding functionalities; a first binding functionality towards the target and a second binding functionality towards a catching ligand comprised in a solid support; and (ii) the first binding functionality comprises at least two binding sites for the target, and the target comp&#xac; rises at least two binding sites for the TBP.

Method for purification of target polypeptides

The invention provides compositions and methods for the detection of targets in a sample; in particular, an in situ hybridization (ISH) sample. Probes and detectable labels may be provided in multiple layers in order to increase the flexibility of a detection system, and to allow for amplification to enhance the signal from a target. The layers may be created by incorporating probes and detectable labels into larger molecular units that interact through nucleic acids base-pairing, including peptide-nucleic acid (PNA) base-pairing. Optional non-natural bases allow for degenerate base pairing schemes. The compositions and methods are also compatible with immunohistochemistry (IHC), immunocytochemistry (ICC), flow cytometry, enzyme immuno-assays (EIA), enzyme linked immuno-assays (ELISA), blotting methods (e.g. Western, Southern, and Northern), labeling inside electrophoresis systems or on surfaces or arrays, and precipitation, among other general detection assay formats. The invention is also compatible with many different types of targets, probes, and detectable labels.

In situ hybridization detection method

Immunohistochemistry detection method

An apparatus for processing at least one biological sample accommodated on at least one carrier member () in a chamber includes, at least one reservoir () able to accommodate a fluid on a surface inside the chamber adjacent to and/or facing a substantial part of the at least one biological sample. The apparatus may comprise a bottom member () arranged to support at least one carrier member () carrying at least one biological sample and a lid () including at least one fluid reservoir (). The reservoir filled with water provides humidity to the chamber and impedes drying out of the sample.

Apparatus and method for processing biological samples and a reservoir therefor

There is disclosed a method of pre-treatment and staining, according to a protocol, of a biological sample disposed upon the surface of a carrier, the method comprising the step of recording at least one parameter relating to at least one protocol step in a non-volatile memory located either upon or within the carrier or a device incorporating the carrier. Also disclosed is a device comprising: a non-volatile memory; a surface of the device adapted to carry a biological sample; and communications means electrically coupled to the memory for enabling data transmission to or from an external apparatus. Also disclosed is a method of controlling processing of a biological sample disposed upon a carrier, comprising: providing, upon or within the carrier or an apparatus holding the carrier, a non-volatile memory having information relating to sample processing priority or protocol; reading the information; and scheduling the processing based upon the information.

Method of pre-treatment and staining of a biological sample and device for support of a biological sample and methods of using such device

The present invention concerns an apparatus for automatic processing at least one biological sample accommodated on a carrier member, such as a slide by applying a predetermined amount of reagents in a predetermined sequence according to a processing protocol, said apparatus comprising; a housing frame; at least one processing section for accommodating at least one slide, the at least one processing section is provided within the housing; a hood cover protecting the at least one processing section in said housing; wherein the hood cover completely encloses the processing section defining an interior space; and wherein the apparatus further comprises climate control device provided to control the environment within the interior space.

Apparatus and methods for processing biological samples and a reservoir therefor

The invention relates to methods and compositions of detecting a biological marker in a sample. In certain embodiments, the methods and compositions comprise a first binding agent that specifically binds to a biological marker in a sample; a second binding agent linked to a first polymer, wherein the second binding agent specifically binds to the first binding agent; a third binding agent linked to a second polymer, wherein the third binding agent specifically binds to the second binding agent; and a detectable substance. In certain embodiments the biological markers are detected immunologically.

Methods and compositions for immuno-histochemical detection

The inventions relates to compositions and method for determining the absolute counts of cells per unit volume of a sample. Such a method comprises: (a) providing a container containing (i) a predetermined quantity of microparticles; and (ii) a cell-binding agent; in which the microparticles are disposed in or on a matrix which adheres to at least one wall of the container such that substantially all the microparticles are thereby attached to the container; (b) adding a known volume of sample to the container; (c) determining the ratio of microparticles to cells by counting microparticles and cells in a volume of the sample; and (d) determining the absolute count of cells by multiplying the number of cells per microparticle by the concentration of microparticles in the sample. Preferably, the matrix retains substantially all the microparticles in or on the container during routine handling, including inversion, of the container, in the absence of mechanical retaining means such as a retainer grid in the container.

The present invention relates to a method for enhancing immunoreactivity of a tissue or cell sample fixed in a fixing medium, a target retrieval composition and its use. The method comprises providing a carrier in a horizontal position, said carrier having thereon a tissue or cell sample, said tissue or cell sample being on top of the carrier; contacting substantially the tissue or cell sample side of the carrier with a buffered target retrieval solution, wherein the target retrieval solution remains otherwise exposed to the environment; heating the tissue or cell sample and the target retrieval solution to a temperature above 100&#xb0; C. The invention furthermore concerns the automated immunohistochemical staining of said samples, in particular the reactivation of antigens masked by fixation.
