The cystic fibrosis gene and its gene product are described for both the normal and mutant forms. The genetic and protein information is used in developing DNA diagnosis, protein diagnosis, carrier and patient screening, drug and gene therapy, cloning of the gene and manufacture of the protein, and development of cystic fibrosis affected animals.

Cystic fibrosis gene

The invention consists of episomal expression cassettes for expression of a transgene in gene therapy. The expression cassettes consist of regulatory elements of the human cytokeratin gene and a transgene. The invention also includes of liposomes for transfection of epithelial tissue with the cassettes in treatment of cystic fibrosis, emphysema, cancers of epithelial origin arising in the lung or other organs.

A rapid and sensitive detection method capable of reliably distinguishing fragments identical to a defined sequence from fragments containing a single base pair mismatch relative to the defined sequence. The method employs a peptide analog antisense probe, such as a peptide nucleic acid (PNA) probe, which hybridizes with DNA to forms complexes having higher thermal stability, but greater sensitivity to base pair mismatches, than corresponding DNA/DNA complexes. The method may be used in a number of applications, including genetic screening and hybridization sequencing applications.

A modified DNA sequence encoding full length cystic fibrosis transmembrane conductance regulator protein is provided to facilitate propagation and/or expression of the protein in living cells and in particular, bacterial cells. The modified DNA sequence comprises at least one of the 13 base pair repeat of exon 6b of the normal gene encoding the conductance regulator protein, as one or more normal nucleotides of the 13 base repeat substituted with an alternate nucleotide which, however, continues to code for the corresponding normal amino acid. Methods of detecting cystic fibrosis gene by nucleic acid hybridization

A method is provided for detecting, ordering and mapping genes or DNA sequences in the genome of eukaryotic cells. The method comprises the steps of releasing free chromatin from the nuclei of the cells and contacting the released free chromatin with at least one detectable probe capable of hybridizing to the genes or DNA sequences to be detected, thereby rendering the genes or DNA sequences detectable.
