The present disclosure relates to compounds, complexes, compositions, kits and methods for determining interacting and/or binding sites between e.g., proteins, proteins and nucleic acids, proteins and small molecules, or intrachain protein domains. The disclosure provides rapid and direct positive identification of the contact interface region between such molecules, and can be applied to individual interacting pairs, as well as large-scale or global interactions.

Binding domain mapping and compounds, compositions, complexes, methods, and kits related thereto

A liquid including an organic component, a stabilization component, and a preservation component is provided. The organic component, the stabilization component, and the preservation component are different components. The organic component present in the liquid includes a volatile organic solution with a refractive index of 1.5 or below. Further, the liquid is a visualization solution that provides a resolving power of a lens by 1/1.5.

A wearable device measures, tracks, and monitors a wearer's physical physiological conditions during a rehabilitation period. Metrics such as temperature, patellar shifting, limb circumference, and acceleration may be collected. Changes in monitored conditions may be assessed for detecting medical abnormalities which require specialized attention, including for example embolisms or infections. A collapsible sample collection device with a novel, collapsible 3D capture open affinity net dispersed throughout the volume of the container, said net with tethered affinity moieties, is designed to address the need for low-cost, simple harvesting and concentration of diagnostic analytes that generally does not require any ancillary equipment. The collection device generally permits analytes of interest to be efficiently harvested from body fluid samples, in one step, concentrated, collapsed into a flat mailing size, and protected from degradation without the need for traditional cold chain technologies. The concentration factor will increase sensitivity by several orders of magnitude when coupled with any type of downstream analyte measurement, including point of care testing. The collection device, in various embodiments, obviates the need for handling transport or storage of the fluid portion of the body fluid sample (e.g. blood, urine, saliva, etc.) with a volumetric concentration factor of 5 to 100 fold. The collapsed collection device generally may be mailed to a laboratory for analysis via any chemical, immunological, electrical or other analytical methods.

Capture particles for harvesting analytes from solution and methods for using them are described. The capture particles are made up of a polymeric matrix having pore size that allows for the analytes to enter the capture particles. The pore size of the capture particles are changeable upon application of a stimulus to the particles, allowing the pore size of the particles to be changed so that analytes of interest remain sequestered inside the particles. The polymeric matrix of the capture particles are made of co-polymeric materials having a structural monomer and an affinity monomer, the affinity monomer having properties that attract the analyte to the capture particle. The capture particles may be used to isolate and identify analytes present in a mixture. They may also be used to protect analytes which are typically subject to degradation upon harvesting and to concentrate low an analyte in low abundance in a fluid.

An assay device for determining the presence of analytes in a cell lysate comprises a porous support member and a plurality of binding reagents arranged and immobilized at multiple reaction sites on the support member. The binding reagents are selected and arranged to assess the status of a selected cellular signal transduction pathway/protein-protein interactive network. In a further aspect, a method for assessing the status of a signal transduction pathway comprises generating a lysate of cells, the lysate retaining one or more pathway molecules present in one or more states and the pathway molecules reflecting signal transduction events taking place in the cells. The method further includes applying the lysate to an immobilized series of binding reagents which can discriminate the pathway molecules and their states. Binding events between the pathway molecules and the binding reagents are identified and the state of the selected signal pathway is determined.
