An apparatus and method for packaging of an optical sensing fiber is disclosed. The apparatus includes a substrate with a plurality of openings, and each opening is configured for holding an optical sensing assembly. The assembly is positioned in the opening with a tip of the assembly extending through the opening to be suspended from the substrate. In addition, openings are arranged so the assembly positioned therein avoids contacting another assembly positioned therein. The apparatus can include a support member for supporting the substrate and positioning the substrate so the tip of the assembly suspended from the opening in the substrate contacts solution in one of a plurality of wells in a container adjacent to the substrate. The assembly can be configured for preparing of the optical assembly for assay. An agitation assembly for agitating the container to create flow of the solution in the container wells over an optical sensing assembly is also disclosed.

Apparatus and method for detecting the presence or amount or rate of binding of an analyte in a sample solution is disclosed. The apparatus includes an optical assembly having first and second reflecting surfaces separated by a distance 'd' greater than 50 nm, where the first surface is formed by a layer of analyte-binding molecules, and a light source for directing a beam of light onto said first and second reflecting surface. A detector in the apparatus operates to detect a change in the thickness of the first reflecting layer resulting from binding of analyte to the analyte-binding molecules, when the assembly is placed in the solution of analyte, by detecting a shift in phase of light waves reflected from the first and second surfaces.

Apparatus and method for detecting an analyte in a sample based on optical interference. The apparatus includes a light source, detector unit and one or more disposable detector tips. The apparatus also includes an optical coupling assembly that couples light from the source to the detector tips, and from the detector tips to the detector unit.

Disclosed are enzyme assays using biolayer interferometry. Assays may be carried out using immobilized substrate or with a substrate capture format. In certain embodiments, the assays are carried out using unlabeled substrates. The methods are broadly applicable to enzyme assay measurements, can be carried out in vivo or in vitro, and are easily multiplexed.
