Human papillomavirus (HPV) protein expression is downregulated in patients infected with HPV by administration of PPAR&#x3b3; ligand, selective inhibitor of cyclooxygenase-2 (COX-2), diaryl heterocycle, inhibitor of HPV protein from a natural source and/or certain non-steroidal anti-inflammatory drugs.

Treatment of HPV caused diseases

Cancers associated with overexpression of HER-2/neu are treated with a selective inhibitor of cyclooxygenase-2 as the sole treating agent or said inhibitor in combination regimen with HERCEPTIN&reg; and/or standard therapy. Uses include adjuvant therapy for HER-2/neu positive breast cancer and treatment of HER-2/neu positive breast cancer that has metastasized.

Treating cancers associated with overexpression of HER-2/neu

Cancer associated with overexpression of class I family of receptor tyrosine kinases, e.g., with overexpresssion of HER-2/neu or overexpression of epidermal growth factor receptor, are treated with strongly binding PPAR&ggr; ligands.

Treating cancers associated with overexpression of class I family of receptor tyrosine kinases

Cells are transfected with a construct containing transcriptional promoter element(s) that have been implicated in carcinogenesis or inflammation ligated to a reporter gene. Determination of inhibition of activation of said promoter element(s) by putative agent indicates the agent is a candidate as a drug or source of a drug for prophylaxis or treatment of cancer or inflammation. The method has particular application to screening agents as candidates for drugs or sources of drugs for prophylaxis or treatment of human disorders caused or mediated by cyclooxygenase-2 and/or matrix metalloproteinases.
