HUMAN-DERIVED BACTERIA THAT INDUCE PROLIFERATION OR ACCUMULATION OF REGULATORY T CELLS

It was found that bacteria belonging to the genusinduce accumulation of regulatory T cells (Treg cells) in the colon. Moreover, the present inventors found that regulatory T cells (Treg cells) induced by from these bacteria suppressed proliferation of effector T-cells. From these findings, the present inventors found that the use of bacteria belonging to the genusor a physiologically active substance derived therefrom made it possible to induce proliferation or accumulation of regulatory T cells (Treg cells), and further to suppress immune functions.

Composition for inducing proliferation or accumulation of regulatory T cells

Disclosed herein are bacteriophage compositions and therapeutic uses thereof. The disclosure also relates to bacteriophage that are capable of lysingbacterial strains, e.g., strains that are associated with inflammatory bowel disease, and thereby capable of modulating disease.

Bacteriophage for modulating inflammatory bowel disease

The result of orally administering saliva derived from a Crohn's disease patient or an ulcerative colitis patient to germ-free mice has revealed that Th1 cells markedly increased in the colons. Further, from the bacterial microbiota in the intestines of the mice in which such an increase in Th1 cells were observed, bacteria have been successfully isolated which caused strong Th1 cell induction in the colon upon intestinal colonization.

Bacterium capable of inducing TH1 cells

With the aim of proving an antibacterial composition against oral bacteria and the like capable of inducing Th1 cell proliferation or activation in an intestinal tract, the present inventors have found out that bacteria that suppress colonization and the like of the oral bacteria and the like in the intestinal tract are present in an intestinal microbiota. Moreover, the present inventors have succeeded in isolating intestinal bacteria that suppress intestinal colonization and the like of oral bacteria and the like.

Anti-bacterial composition against TH1 cell-inducing bacteria

Strains of human-derived bacteria have been obtained from complex fecal samples and shown to induce accumulation of Th17 cells in the intestine and promote immune functions. Pharmaceutical compositions containing these bacteria can be used as anti-infectives and as adjuvants in mucosal vaccines.

Compositions and methods for induction of Th17 cells

Provided herein are compositions and methods for the induction and/or proliferation of CD8+ T-cells. The disclosure also provides methods of treatment of diseases that can be treated by the induction and/or proliferation of CD8+ T-cells.

Compositions and methods for the induction of CD8+ T-cells

Provided herein are methods of treating graft-versus-host disease in a subject by administering pharmaceutical compositions containing bacterial strains of the Clostridia class. Also described herein are exemplary human-derived bacteria belonging to the Clostridia class, combinations of which have been shown to induce accumulation of regulatory T cells (Treg cells) in the colon and suppress immune functions, and are therefore useful for mitigating pathological immune responses. Pharmaceutical compositions containing these and/or related bacteria can be used to prevent and treat immune-mediated diseases such as graft-versus-host disease.

Human-derived bacteria that induce proliferation or accumulation of regulatory T cells

Species of human-derived bacteria belonging to the Clostridia class have been shown to induce accumulation of regulatory T cells (Treg cells) in the colon and suppress immune functions. Pharmaceutical compositions containing these bacteria can be used to prevent and treat immune-mediated diseases such as autoimmune diseases.
