In a liquid chromatograph, the disulfide adsorption of iron atoms in a filter for a column is avoided and thus peak tailing in a chromatogram is prevented to thereby prevent a lowering in the sample recovery rate. A liquid chromatograph column comprising a hollow tubular column body, a bead-shaped packing material which is packed in the hollow part of the column body, a filter provided with a mesh which has a mesh size smaller than the particle diameter of the packing material, and a cap which presses the filter against the packing material to thereby enclose the packing material within the column body, characterized in that the main component of the filter is nickel or hastelloy.

It is possible to correctly determine whether a change in the pressure or flow rate is caused by normal opening and closing operations of a check valve and to monitor in real time whether an operation of a check valve is normal or abnormal in a liquid supply device. Light is introduced in the check valve and a change in the quantity of light transmitted through or reflected by the check valve, caused by the opening and closing of the valve is detected, so that the opening and closing operations of the check valve can be directly detected. The check valve is arranged in a pipe in the liquid supply device, and a change in the pressure in the pipe is monitored on the basis of a signal from a pressure sensor and a signal which represents the opening and closing of the check valve.

Both a column having high-pressure resistance and capable of performing high-speed separation and analysis even with a small flow rate and a liquid chromatograph apparatus using the column are provided. A separation column according to the present invention has a monolithic rod being arranged in the center, being made of a porous material, and having a circular cross section, a filler layer arranged such that it encircles a circumferential surface of the monolithic rod, and a cylindrical support medium arranged outside the filler layer. The filler layer is formed by filling a tube-like gap between the monolithic rod and the support medium with particles or beads.

After various sugar nucleotide solutions and glycosyltransferases (or primers) have been mixed, they are introduced into a reaction tank (column) with primers (or glycosyltransferases) immobilized thereon. Then solutions coming out of the reaction tank are led to an ultrafiltration column. The oligosaccharide synthesizer according to the present invention is equipped with a flow path for ensuring that glycosyltransferases or primers separated by the ultrafiltration column are returned into a container for storing each solution in a sample injector.

A chromatographic analyzer is provided for facilitating curve fitting by means of the linear least-square method for a chromatogram that contains a plurality of overlapping peaks. A chromatographic data processor executes fitting processing to each peak in an arbitrary time region having the plurality of peaks of the chromatogram starting from the front or back side of the time region or from the back side of the time region, and the processed peaks are subtracted from the chromatogram in the time region so that the plurality of peaks in the chromatogram can be separated from one another.

A method for analyzing a multi-channel chromatogram is realized for accurately resolving overlapping peaks on a multi-channel chromatogram to permit analysis of the composition of a sample. First, components of overlapping peaks are specified. A data matrix Dij is determined, and compressed in the wavelength direction to obtain a data matrix vector Di. Next, the two-dimensional data is deconvoluted, final standard deviation sis registered, and the deconvoluted two-dimensional data is reconvoluted to a retention intensity matrix. A quantitative spectral intensity matrix is computed, components are identified, and the identified components are quantified. Subsequently, an eigenvalue problem is solved, and the number n of components is estimated. The deconvolution and computation of the eigenvalue problem are iteratively executed until the peaks are isolated. Then, elution profiles are calculated, the components are identified, and the identified components are quantified.

Method and apparatus for analyzing multi-channel chromatogram

The present invention provides a sugar chain synthesizer capable of continuously reacting sugar chains when a plurality of sugar chains are successively reacted. The sugar chain synthesizer of the present invention includes a plurality of vessels containing respective sugar nucleotide solutions, a plurality of vessels containing respective glycosyltransferases, and a reactor containing a primer that is a water-soluble polymer, into which the above described sugar nucleotide solution and glycosyltransferase are introduced. In the present invention, components in a reaction solution obtained in the reactor are separated through an ultrafiltration column, and a reaction product is then returned to the above described reactor, so as to continuously synthesize sugar chains. Although it is a complicated synthesis of sugar chains, it becomes possible to carry out such synthesis continuously and automatically.

Methods and instruments for identification of glycosylated proteins and peptides

The present invention provides a sugar chain purification instrument which can continuously and efficiently purify the sugar chain. According to the present invention, the instrument includes an immobilized protease column in which a carrier which can be packed in the column and on which protease is immobilized is packed and an immobilized glycopeptidase column in which a carrier which can be packed in the column and on which glycopeptidase is immobilized is packed, these columns being connected in series, and the reaction is carried out while continuously passing a glycoprotein to be decomposed together with a suitable buffer solution through the above columns. The step of liberation of sugar chain from the glycoprotein is highly simplified and labor-saving, and liberation of the sugar chain can be moderately and simply performed.

An object of the present invention is to provide a mass spectrometry system capable of improving an efficiency of obtaining information on a structure of substances, shortening a time taken for a measurement and substance identification, and improving identification accuracy. The system comprises: a process of mass analyzing an ionized analyte; a first fragmentation process where a first ion is selected from the ions observed in a mass spectrometry to fragment it; a process of mass analyzing a plurality of the ions generated in the first fragmentation process; a process of determining fragment ion combination capable of reconstructing the first ion using a result of the mass spectrometry; a second fragmentation process where the fragment ions contained in the fragment ion combination are fragmented; and a process of mass analyzing the fragment ions generated in the second fragmentation.

Method of identifying substances using mass spectrometry

The invention intends to determine a presence ratio between mixed compounds which are difficult to separate from each other by a separation unit and cannot be discriminated by MS. In a mass spectrometer with an MSanalysis function for ionizing a sample eluted from a separation unit for separating the sample into individual components, fragmenting an ion of a desired mass number, and performing a mass analysis of fragment ions, the mass spectrometer includes a database storing correlation information between an isomer presence ratio and a particular ion intensity ratio in a mass spectrum per isomer. An isomer ratio can be clarified even for a mixed sample of isomers, such as enantiomers, which are difficult to separate from each other by the separation unit and cannot be discriminated by MS.
