A clearing pretreatment method of a biological sample having a size of at most 1 mm, that is, a spheroid or organoid, according to the present invention uses a phosphate-buffered saline (PBS) rather than a conventional sucrose solution as a pretreatment solution, thus solving the problem in which the spheroid or organoid floats above the surface of water due to the density difference such that the structure of the sample is damaged. Therefore, there is an effect in that the spheroid or organoid can be transparentized while maintaining the original shape thereof, thus making it possible to image deep parts.

The present invention relates to a method for fabrication of a three-dimensional lung organoid comprising human stem cell-derived alveolar macrophages. Specifically, a lung organoid is fabricated by co-culturing cells not expressing the definitive endoderm marker CRCX4 according to a fabrication method of the present disclosure. The lung organoid comprises type 1 and type 2 alveolar epithelial cells as well as alveolar macrophages and realizes infectious or inflammatory responses unlike conventional lung organoids that contain no immune cells and as such, can be advantageously used in studying mechanisms of related lung diseases, excavating biomarkers, developing therapeutic agents, and so on.

The present invention relates to a heart targeting agent comprising tannic acid, in which the agent aids a heart disease therapeutic drug to be delivered to the heart and bind to the cardiac myocardium by inducing tannylation of the drug so as to make heart targeting and accumulation of the drug possible. Unlike the conventional invasive method used for the traditional drugs to be able to target the heart, the agent of the present invention can help a drug to target the heart with high efficiency just via non-invasive intravenous administration.

The present invention relates to a composition for immunostaining cleared large tissues and a method for immunostaining cleared large biological tissues using the same. The composition for immunostaining cleared large tissues increases antibody permeability in cleared biological tissues, in particular, large biological tissues having a thickness of 3 mm or more, to allow antibodies to penetrate deep into thick tissues, thereby overcoming immunostaining problems associated with limited permeation of antibodies in existing methods, reduces the time required for immunostaining large tissues, thereby enabling simple and rapid three-dimensional bioimaging with high resolution for large biological tissues, and may be beneficially used, through structure images, for identifying the causes of various diseases, developing new treatment methods, and predicting the efficacy and toxicity of drugs. The present invention relates to a composition for clearing biotissue and a clarity method for biotissue using the same. The composition for clearing biotissue and the clarity method for biotissue using the same of the present invention do not need a high price electrophoresis apparatus and expensive solutions, and can not only be applied to various biotissues including brain, liver, lung, kidney, intestine, heart, muscle, and blood vessel, without damaging any of them but also prevent bubble formation, color change, and dark sediment; improve the clarity of biotissues; and help antibody-staining of the cleared tissues. Therefore, it is useful to identify the reason of various diseases through structural imaging of biotissue and to establish a treatment method.
