This invention relates to a method to produce gene alterations in the genomes of a eukaryotic and prokaryotic host cell. The method comprises utilizing Argonaute from Natronobacterium gregoryi (NgAgo) or a mutant thereof, and complementary 5' phosphorylated single-stranded DNA that target the enzyme to cleave specific regions of the chromosome. Additionally, N-terminal truncations (deletion of the repA domain; N-del), or its mutants including N-del/E598A, N-del/D601P, and N-del/E602P reduces random cleavage and can be used for targeted gene editing with a guide DNA. An expression system or a host cell and method of creating thereof are also in the scope of this application.

NgAgo-based gene-editing method and the uses thereof

Provided herein are novel proteins and protein domains from newly discovered anaerobic fungal species. The anaerobic fungal species have unique enzymatic capabilities, including the ability to digest diverse lignocellulosic biomass feedstocks and to synthesize secondary metabolites. The scope of the invention encompasses novel engineered proteins comprising glycoside hydrolase enzymes, dockerin domains, carbohydrate binding domains, and polyketide synthase enzymes. The invention further encompasses artificial cellulosomes comprising novel proteins and domains of the invention. The scope of the invention further includes novel nucleic acid sequences coding for the engineered proteins of the invention, and methods of using such engineered organisms to degrade lignocellulosic biomass and to create polyketides.

Proteins from anaerobic fungi and uses thereof

Methods and nucleic acid sequences for the synthesis of biotemplates in a non-plant based expression system are provided. Such biotemplates include Barley stripe mosaic virus viral-like particles (BSMV-VLPs) that are capable of self-assembly due to being operatively linked with an origin of self-assembly with the Barley stripe mosaic virus capsid protein (BSMV-CP). Also provided are BSMV-VLPs that are capable of self-assembly due one or more site-directed mutations on the BSMV-CP, and BSMV-VLPs that exhibit enhanced stability due to such site-directed mutation(s).

Nanoparticles and biotemplates with tunable length and methods of manufacturing the same

This present patent application relates to a method for controlling a targeted gene expression with an environmental trigger useful for manufacturing a natural product or an analogue thereof biologically. In particular, the present invention discloses an expression system comprises a gene expressing for a fusion protein of elastin-like polypeptides (ELPs) and a transcription factor, wherein a targeted gene expression is regulated by the phase change of the fusion protein initiated by an environmental trigger, including changes of temperature, pH value, ionic strength or a combination thereof. The method, the expression system and their products are within the scope of this invention.
