The present disclosure provides a method of treating a human patient comprising the steps of: systematically administering multiple doses of a parenteral formulation of a replication capable oncolytic adenovirus of subgroup B in a single treatment cycle, wherein the total dose given in each dose is in the range of 1×10to 1×10viral particles, and wherein each dose of virus is administered over a period of 1 to 90 minutes, for example at a rate of viral particle delivery in the range of 2×10particles per minute to 2×10particles per minute. The disclosure further extends to formulations of the oncolytic adenoviruses and combination therapies of the viruses and formulations with other therapeutic agents.

Dosing regime and formulations for type B adenoviruses

A modified adenovirus, in particular Enadenotucirev (EnAd), armed with at least two bispecific T cell engagers each comprising at least two binding domains, wherein at least one of the domains is specific for a surface antigen on an immune cell of interest, such as a T-cell of interest. Also provided are a composition, such as a pharmaceutical formulation comprising the virus, use of the virus and virus formulations for treatment, such as in the treatment of cancer. The disclosure also extends to processes for preparing the virus.

The present disclosure relates to a group B adenovirus comprising a sequence of formula (I): 5'ITR-B1-BAB2-BX-BB-BY-B3-3&#x2032;ITR wherein: B1 is bond or comprises: E1A, E1B or E1AE1B; BA comprises: E2B-L1-L2-L3-E2A-L4; B2 is a bond or comprises: E3; BX is a bond or a DNA sequence comprising: a restriction site, one or more transgenes or both; BB comprises L5; BY is a bond or a DNA sequence comprising: a restriction site, one or more transgenes or both; B3 is a bond or comprises: E4; wherein at least one of BX or BY is not a bond, pharmaceutical compositions comprising the same and use of the viruses and compositions in treatment, particularly in the treatment of cancer. The disclosure also extends to plasmids and processes employed to prepare the said viruses.

Modified viral particles wherein the viral particles, typically adenoviral particles, are modified by glycosylation and the use of the modified viral particles to deliver heterologous nucleic acid to cells. Also disclosed are pharmaceutical compositions comprising the same and method of treatment using the same.

Modified virus

A method of modifying the biological and/or physicochemical properties of biological elements such as viruses and other micro-organisms is disclosed in which the biological element is modified by providing it with a coating of a multivalent polymer having multiple reactive groups. This modification can enable some biological elements to be targeted or re-targeted to particular sites in a host biological system and can be useful in connection with viral vectors for gene therapy or antitumor therapy. In other cases the modification can be useful for enhancing or improving the efficiency of viruses or bacterial micro-organisms used for example in pest control, degradation and dispersal of oil deposits and various other industrial, environment or medical applications.
