The present invention is directed to affinity chromatography devices that separate a targeted protein or antibody from an aqueous mixture containing the targeted protein or antibody. The chromatography device may contain a stacked membrane assembly or a wound membrane assembly. The membrane assemblies include (1) at least one polymer membrane that contains therein inorganic particles and (2) at least one impermeable layer (e.g., a thermoplastic polymer in a solid state). The polymer membrane and/or the inorganic particles have an affinity ligand bonded thereto. The affinity ligand may be a protein, an antibody, or a polysaccharide that reversibly binds to the targeted protein or antibody. The chromatography device may be repeatedly used and may be cleaned with a caustic solution between uses. The chromatography devices has a dynamic binding capacity (DBC) of at least 30 mg/ml (or 0.07 micromol/ml) at 10% breakthrough at a residence time of 20 seconds or less.

Affinity chromatography devices

A filtration article including a composite filtration structure, a seal region positioned within the composite filtration structure, and an attachment weld that interconnects the seal region and an outer casing is provided. The composite filtration structure includes a stacked filter material including at least two fluoropolymer membranes and a support layer. The support layer may be formed of a thermoplastic or thermoset polymer. When heated, the polymer in the support layer melts and flows into the fluoropolymer membranes. In exemplary embodiments, the polymer is imbibed through the thickness of the fluoropolymer membranes. The attachment weld may be positioned at any location within the seal region so long as the attachment weld is positioned a distance from the outer periphery of the composite filtration structure. The filtration article is small in size, is lightweight, and may be used as a sterilizing grade filter. Methods of producing the filtration article also provided.

Fluoropolymer article for bacterial filtration

A V-panel filter apparatus and method of construction are provided. The method includes flowing curable potting material into a recess portion of an end cap, positioning first end edge surfaces of first and second filter panels into the contained potting material, and curing the potting material with the first and second filter panels positioned to diverge away from the end cap to define a V-shaped configuration. The method may include flowing curable potting material into a recess portion of a first side plate, positioning edge surfaces of the first and second filter panels into the contained potting material, and curing the potting material. Such steps may be repeated in relation to a second side plate and a header member. By utilizing a pot-and-place sealing methodology, improved seal interfaces are realized. Stability and construction advantages may be provided by snap-fit features of the header member and first and second side plates.
