A system and method are shown for performing cool, on-column sample injection into a chromatographic device having a capillary column which injection is achieved through the use of a syringe having a needle. The column diameter is minimized by the provision of an inlet assembly connected to the inlet end of the column, a sealing member disposed in the inlet assembly, for forming a fluid tight seal in the inlet end when the sample is injected onto the column to prevent the sample from escaping and for reducing the effective length of the needle at the point of insertion of the needle into the column and a guide member for reducing the effective length and for supporting the needle during insertion into the column. A temperature controller for controlling the temperature of the column in the region where the sample is injected is also disclosed. The sealing member is shown to include an elastomeric member and further the system includes a mechanism for continuously cooling the elastomeric member. The elastomeric member is shown to include a septum having a bore formed therein for passage of the needle therethrough. A new sample vial closure is also shown for use in the system, which closure applies only a minimal axial force on the needle. In such a system and method the need for a long syringe is obviated. The column inner diameter can be minimized by the system and method which permits the needle of the syringe to having a minimum outer circumference.

Apparatus and methods are disclosed for performing a chromatographic separation of a given compound utilizing programmed temperature and mass flow wherein the compound is injected into a pressurized carrier gas and passed through a column and wherein a portion of the column is subjected to a temperature profile, by determining the pressure of the carrier gas and generating a pressure information signal representative of such pressure, storing desired carrier gas mass flow information and temperature profile information, and controlling the pressure of the carrier gas in response to the mass flow information, the temperature profile information and the pressure information signal, so that as the temperature profile information changes with time, the desired mass flow of carrier gas at a corresponding time is maintained by controlling the pressure of the carrier gas.

Apparatus and methods are disclosed for performing a chromatographic separation of a given compound wherein the compound and a carrier gas are passed through an injection port and onto a column and wherein a portion of the column is contained in an oven. The apparatus and method are shown to include a first heater for heating the injection port in response to a first control signal, a first temperature sensor for sensing the temperature of the injection port and for generating a first temperature signal representative of the temperature of the injection port, a second heater for heating the oven in response to a second control signal, a second temperature sensor for sensing the temperature of the oven and for generating a second temperature signal representative of the temperature of the oven and a cooling member for simultaneously cooling the injection port and the column. The cooling member can include various conduit arrangements for directing a flow of cryogen fluid. There is also shown a valve for controlling the flow of cryogen fluid through the cooling member in response to a third control signal, and a control member for receiving the first and second temperature signals, and for generating the first, second and third control signals in relation to the first and second temperature signals, so that the temperature in the injection port and the oven are maintained at a desired level.
