An observation apparatus includes a display apparatus that displays a display pattern, a display projection optical system that projects a light beam from the display apparatus, and forms an image of the display pattern, a combining optical element that combines a light beam from a sample and a light beam from the display apparatus, and an eyepiece optical system that enables an observer to simultaneously observe an image of the sample and an image of the display pattern, in which a numerical aperture (NA) of a light beam from the display apparatus is smaller than a maximum value of an NA of a light beam from the sample, and is larger than a minimum value of an NA of a light beam from the sample, at a position of an image on an optical path that is formed after light beams are combined by the combining optical element.

A microscope includes a light source; a condenser lens irradiating a sample with a light from the light source; an objective facing the condenser lens across the sample; a first polarization plate placed between the light source and the condenser lens; a condenser turret placed between the first polarization plate and the condenser lens and having a plurality of optical elements placed inside; a polarization plate placed on the image side with respect to the objective; and a compensator placed between the first polarization plate and the polarization plate. An immersion microscope objective formed of thirteen or fewer lens elements includes, in order from the object side, first and second lens groups of positive refractive power, a third lens group, a fourth lens group having negative refractive power with its image-side surface being concave, and a fifth lens group having positive refractive power with its object-side surface being concave. The first lens group includes, in order from the object side, a lens component that consists of a lens element of positive refractive power (when computed as being in air) and a meniscus lens element having its concave surface on the object side. An immersion microscope objective includes, in order from the object side, a first lens group having positive refractive power for converting the luminous flux from an object into convergent luminous flux, a second lens group having the refractive power lower than that of the first lens group, and a third lens group, and satisfies the following conditional expression where NA indicates the numerical aperture on the object side, and dindicates a working distance:3 mm&#x3c;NA&#xd7;0&#x3c;8 mm.

A microscope includes a laser light source, an optical system which changes a beam diameter, and a field stop disposed at a position conjugating with a sample plane, in this order beginning from the side of the laser light source. Provided is a scanning laser microscope including a first laser light source for emitting ultrashort pulsed laser light; a scanning unit for two-dimensionally scanning the ultrashort pulsed laser light on a specimen; a second laser light source for emitting continuous laser light; an irradiation-position adjusting unit for performing two-dimensional adjustment of an irradiation position of the continuous laser light on the specimen; an objective lens for focusing the ultrashort pulsed laser light and the continuous laser light onto the specimen and for collecting fluorescence generated in the specimen; a light-detecting unit for detecting the fluorescence, which is split off from a light path between the objective lens and the scanning unit; and a continuous-laser-light switching unit for permitting radiation of the continuous laser light when the light-detecting unit is not detecting required fluorescence from the specimen on the basis of the ultrashort pulsed laser light.

Immersion type microscope objective

A total internal reflection fluorescence microscope includes a light source which generates laser light having a plurality of wavelengths, a focal optical system which irradiates the laser light from the light source on a specimen at a predetermined incident angle via an objective lens and which generates evanescent illumination, a fluorescence observation unit which observes fluorescence generated from the specimen due to the evanescent illumination, an incident angle adjuster which adjusts the incident angle of the laser light irradiated on the specimen, and a controller which controls the incident angle adjuster such that the amount of the permeation depth of the evanescent light is the same when the wavelength of the laser light from the laser light source is switched.

Total internal reflection fluorescence microscope

A fluorescent microscope comprises a light source, an optical illumination system which forms an optical path to irradiate a specimen with a light beam from the light source, an objective lens which condenses the light beam of the optical illumination system onto the specimen, an optical device which is disposed on the optical path of the optical illumination system and which decenters the light beam by decentering an optical axis of the optical path, and a slit which passes the light beam decentered by the optical device through a total reflection illumination region on an emission pupil surface of the objective lens.

Total reflection fluorescent microscope

An illumination optical system for a microscope is provided wherein the illumination state can be successively changed between Koehler illumination and critical illumination while ensuring one or more conditions are satisfied so that the image of the light source illuminates an appropriately large region of the field of view of the microscope during critical illumination. Preferably, no cemented lens elements are used so that degradation of the cement caused by ultraviolet light sources is avoided, thereby enabling an appropriate illumination type to be provided at will and without degradation of the optical components of the illumination optical system over time.
