Provided are compositions comprising polynucleotides encoding modified diacylglycerol acyltransferase-1 (DGAT1) polypeptides having improved properties, such as increased enzymatic activity and/or increased stability. Plants, plant cells, seed, grain and comprising the polynucleotides are provided which have one or more of increased fatty acid or protein content. Methods of generating the polynucleotides in plant cells include transformation and genetic modification. Methods of employing the polynucleotides in plants, methods for increasing DGAT1 activity in a plant, and methods for increasing fatty acid content or protein content in a plant are provided.

Modified seed oil content by gene editing

Altering protein functional properties through terminal fusions

The invention provides isolated nucleic acids and their encoded polypeptides that are involved in enhancing the essential amino acid content of a plant. The polypeptide may be derived from a protease inhibitor, and more specifically, a chymotrypsin inhibitor. Chymotrypsin inhibitors that may be modified for use in the invention are present in many plant species. Barley ( ) was initially used to obtain the chymotrypsin inhibitor modified for use in the present invention. Other plant species that may be used as a source for chymotrypsin inhibitor for use in the present invention include and . Proteins with increased levels of essential amino acids

The gene encoding a biotin carboxylase subunit of heteromeric ACCase has been isolated and sequenced. The cDNA sequence and deduced amino acid sequence is set forth in FIG.,and has been accorded GenBank Accession No. L38260. By controlling expression of the gene of the present invention, carboxylation of acetyl-CoA to produce malonyl-CoA may be controlled. Thus, by introducing constructs of the gene of the present invention in sense or anti-sense orientation, carboxylation of acetyl-CoA to produce malonyl-CoA may be increased or decreased. Consequently, fatty acid synthesis in plants and seeds which depends on malonyl-CoA in the plastid may also be controlled.
