A drug coating layer on a non-porous nylon elastomer balloon substrate surface comprising a plurality of elongated bodies with long axes wherein each elongated body is formed of a crystal of a water-insoluble drug and is independently formed on the substrate surface, wherein the long axes of the elongated bodies are substantially linear in shape, and the long axes of the elongated bodies form an angle in a predetermined range with respect to a substrate plane with which the long axis of the elongated body intersects. The drug coating layer can provide low toxicity and a high intravascular stenosis inhibitory effect.

A coating composition capable of forming a drug coating layer insusceptible to peeling in the process of delivery of a water-insoluble drug to a target tissue is provided. The coating composition contains the water-insoluble drug and a basic compound which is positively charged at physiological pH. A medical device coated with the coating composition is further provided.

A liposome composition, which is obtained by mixing a water-miscible organic solution in which a phospholipid and cholesterol are contained in a total concentration of 100 to 200 w/v % in a water-miscible organic solvent with a first aqueous phase solution in an amount of 3/1 to 12/1 in terms of volume ratio to the water-miscible organic solution, thereby obtaining an emulsion in which the total concentration of the phospholipid and cholesterol in the resulting mixed phase is 15 to 50 w/v %, followed by subjecting the emulsion to external solution exchange with a second aqueous phase solution, wherein an ion gradient is formed between an aqueous phase in an internal region of a liposome membrane, including the first aqueous phase solution, and an aqueous phase in an external region of the liposome membrane, including the second aqueous phase solution, and a drug can be introduced in a high encapsulation amount.

Long-lasting controlled-release liposome composition and method for producing same

A drug coating layer which is a drug coating layer having a morphological form including a plurality of elongated bodies having long axes that each crystal of a water-insoluble drug independently has on a substrate surface, in which the long axes of the elongated bodies are nearly linear in shape, and the long axes of the elongated bodies form an angle in a predetermined range with respect to a substrate plane with which the long axis of the elongated body intersects. A long-lasting, controlled-release local anesthetic liposome preparation is produced by: providing a liposome composition, which is obtained by mixing, in a water-miscible organic solvent, a first aqueous phase solution with a water-miscible organic solution, in which a phospholipid and cholesterol are contained at a defined total concentration, at a defined ratio by volume thereby obtaining an emulsion wherein a total concentration of the phospholipid and cholesterol in the mixed phase is at 15 w/v % to 50 w/v %, followed by subjecting the emulsion to external solution exchange with a second aqueous phase solution to obtain a liposome composition wherein an ion gradient is formed between an internal-region aqueous phase of a liposome membrane made of the first aqueous phase solution and an external-region aqueous phase of the liposome membrane made of the second aqueous phase solution; and encapsulating a local anesthetic in the internal-region aqueous phase according to a remote loading method.

A treatment device includes: a balloon; a biologically active agent part applied on an outer surface of the balloon; a protective sleeve encircling the balloon and being displaceable proximally; and an interposed member having a self-expanding function arranged between the balloon and the protective sleeve. When the protective sleeve is moved proximally in order for the balloon to be exposed to the inside of a blood vessel, the presence of the interposed member prevents the biologically active agent part from being scraped and peeled off by the inner surface of the protective sleeve.

A coating composition for drug eluting medical devices, which is capable of forming a drug coating layer that is not susceptible to peeling during delivery to a target tissue is provided. This coating composition contains: a water-insoluble drug; at least one selected from the group consisting of hyaluronic acid, alkanoyl hyaluronic acids obtained by substituting at least part of hydrogen atoms in the hydroxyl groups of hyaluronic acid with an alkanoyl group, and salts of hyaluronic acid and the alkanoyl hyaluronic acids; and at least one selected from the group consisting of amino-acid esters and salts thereof. A drug coating layer, a medical device and a method of treatment are also provided.
