A method for detecting an abnormal cell based on gene expression analysis, which is useful for cancer diagnosis, is provided. A gene expression analysis method, comprising: measuring the expression level of a transcript of a human MLH1 gene containing the nucleotide sequence as shown in SEQ ID NO: 1 at the 5'-terminus thereof and the expression level of a transcript of a human MLH1 gene containing the nucleotide sequence as shown in SEQ ID NO: 2 at the 5&#x2032;-terminus thereof, in a biological sample; and comparing the expression levels, thereby detecting a cell positive for microsatellite instability.

Method and kit for detection of microsatellite instability-positive cell

A cDNA fragment participating in the maintenance of smooth muscle differentiation was isolated using a culture system of chicken gizzard smooth muscle cells, the differential display method and the subtracted hybridization method. Using the obtained cDNA sequence as a query, cDNA sequences of Helix Research Institute (Japanese Patent Application No. 2000-118776) were retrieved, and thus, a novel gene 'C-NT2RP3001495' was obtained. The protein encoded by this gene has two WW domains that participate in protein interactions in the N-terminal domain. Evidence suggests that this protein binds to other proteins, and thus regulates the intracellular signal transduction, gene expression, and so on, thereby participating in the maintenance of the differentiation of smooth muscle cells. This protein and compounds regulating the expression thereof are markedly useful in developing drugs for various diseases associated with abnormality in the maintenance of smooth muscle cell differentiation.

The present invention relates to a quick and simple method for genetic testing, particularly for SNP testing, using two kinds of primers which are complementary to a mutant target and a wild-type target, respectively, and different in length or migration speed in electrophoresis. These probes are allowed to hybridize with targets, fluorophore-tagged nucleotides are attached to the primers, and electrophoresis is carried out for discriminatory detection.

According to the method, sensitivity is greatly increased by amplification of the amount of pyrophosphate produced in synthesis of the complementary strands without amplifying the copies of nucleotide sequences.

Method for testing nucleic acid sequence

A method for comparing DNA base sequences by comparing similarities between two amino acid sequences translated from two DNA base sequences, respectively, includes (1) a step of dividing each of the first DNA base sequence and the second DNA base sequence into groups of successive three nucleotides each, translating each of these groups of nucleotides into an amino acid, and thereby obtaining a first amino acid sequence and a second amino acid sequence, and (2) a step of determining similarities between each amino acid of the first translated amino acid sequence and each amino acid of the second translated amino acid sequence in view of nucleotide insertions or deletions in the first and second DNA base sequence and amino acid insertions or deletions in the first and second translated amino acid sequence. The method is repeated by shifting the base sequences one base at a time.

Method and apparatus for electrophoresis separation and detection

A DNA analyzing method which bonds a first oligomer of known base sequence to a DNA fragment obtained by digesting a DNA sample with a restrictive enzyme. The oligomer and DNA fragment are hybridized to other oligomers which have the sequences of all combinations of the types of bases within the length of several bases following the known base sequence. The presence or absence of hybridization or complementary DNA strand extension is determined and identifies the DNA fragment terminal sequence from this result. The DNA fragments are then fractionated and analyzed to determine the sequence. This DNA analyzing method provides an effective analysis of mixtures of long DNAs or DNA fragments.
