Provided are a biomarker for evaluating a cerebral A&#x3b2; accumulation state using an amyloid precursor protein (APP)-derived peptide in a living body-derived sample as an index, and a method for analysis thereof. A living body-derived sample derived from a test subject is measured, and at least one ratio selected from the group consisting of APP669-711/APP672-713 (A&#x3b2;1-42); APP672-709 (A&#x3b2;1-38)/APP672-713 (A&#x3b2;1-42); APP674-711 (A&#x3b2;3-40)/APP672-713 (A&#x3b2;1-42); APP672-710 (A&#x3b2;1-39)/APP672-713 (A&#x3b2;1-42); APP672-711 (A&#x3b2;1-40)/APP672-713 (A&#x3b2;1-42); and APP672-711 (OxA&#x3b2;1-40)/APP672-713 (A&#x3b2;1-42) is calculated, and when each of the ratios of the test subject is higher than each of standard levels which are ratios of a person having normal cognitive function NC&#x2212; who is negative for cerebral A&#x3b2; accumulation, an accumulation amount of cerebral A&#x3b2; of the test subject is determined as larger than an accumulation amount of cerebral A&#x3b2; of the person having normal cognitive function NC&#x2212;.

Disclosed is an antibody having a high inhibitory effect on amyloid fibril formation. An antibody is produced by using a liposome containing a GM1 ganglioside at a predetermined ratio as an immunogen. Thus, the sequences of four types of antibodies each having a high inhibitory effect on amyloid fibril formation can be provided.

It is intended to provide an antibody efficacious in diagnosing, preventing or treating Alzheimer's disease, DNA encoding the antibody, a method of screening a drug and drugs. The amino acid sequence and the gene sequence of the variable region of an antibody, which specifically recognizes a GM1 ganglioside-bound amyloid &#x3b2;-protein occurring in the early stage of &#x3b2;-amyloid fibril formation, are determined. Based on the data of the amino acid sequence and the gene sequence thus obtained, an antibody is designed.
