Described are methods for saccharifying starch-containing materials using a glucoamylase, methods for producing fermentation products, and fermentation products produced by the method thereof as well as methods for increasing starch digestibility in a ruminant using at least one of the glucoamylases described herein.

Glucoamylases and methods of use, thereof

The present disclosure provides polypeptides having aminopeptidase activity and isolated nucleic acid sequences encoding the polypeptides. In some embodiments, the disclosure also provides to nucleic acid constructs, vectors, and host cells comprising the nucleic acid sequences as well as methods for producing the polypeptides. In some embodiments, the present disclosure further provides to methods of obtaining protein hydrolysates useful as flavor improving agents.

Exo-specific amylase polypeptides, nucleic acids encoding those polypeptides and uses thereof

A method of oxidizing a saccharide, by contacting the saccharide with an alcohol dehydrogenase (ADH) enzyme selected from a quinone redox cofactor-dependent ADH, a nicotinamide adenine dinucleotide (NAD) redox cofactor-dependent ADH, a nicotinamide adenine dinucleotide phosphate (NADP) redox cofactor-dependent ADH, and any combination thereof is described. An oxidized saccharide obtainable by the method and products, in particular food products and paper products, containing the oxidized saccharide, are also described.

This invention relates to polypeptides, more specifically amylase polypeptides and nucleic acids encoding these, and their uses e.g. as non-maltogenic exoamylases in producing food or feed products.

Amylase polypeptides

We disclose a food additive comprising a PS4 variant polypeptide, in which the PS4 variant polypeptide is derivable from a parent polypeptide having non-maltogenic exoamylase activity, in which the PS4 variant polypeptide comprises substitutions at the following positions: G121D, 134, 141, 157, 223, 307 and 334 with reference to the position numbering of aexoamylase sequence shown as SEQ ID NO: 1.

Polypeptide

G4-forming amylase (PS4), and variants thereof, advantageously can be used in an enzyme-catalyzed high temperature liquefaction step to produce ethanol from starch, e.g., cornstarch. PS4 produces significant amounts of maltotrioses, which can be utilized byin a subsequent fermentation step to produce ethanol. This property of PS4 advantageously allows ethanol to be produced from liquefacted starch in the absence of a saccharification step. PS4 variants are provided that exhibit improved properties, such as thermostability and/or altered exo-specific and endo-specific amylase activity.

We describe a PS4 variant polypeptide derivable from a parent polypeptide having non-maltogenic exoamylase activity, in which the PS4 variant polypeptide comprises an amino acid substitution at position 307 to lysine (K) or arginine (R), with reference to the position numbering of aexoamylase sequence shown as SEQ ID NO: 1. Preferably, the PS4 variant polypeptide further comprises an amino acid substitution at position 70, preferably G70D. The amino acid at positions 272 and 303 of the sequence of the are preferably histidine (H) and glycine (G).

Variants ofsp. TS-23 strain alpha-amylases exhibit improved enzymatic performance, including increased themostability, reduced calcium dependence, increased washing/cleaning performance, and baking ability. Compositions comprising these variants are useful in methods of starch processing, starch liquefaction, fermatation, starch saccharification, cleaning, laundrying, textile desizing, baking, and biofilm removal.
