Certain peptides containing the glucagon.sub.1-21 peptide chain His-Ser-Gln-Gly-Thr-Phe-Thr-Ser-Asp-Tyr-Ser-Lys-Tyr-Leu-Asp-Ser-Arg-Arg-Al a-Gln-Asp, usable as a medicament or a diagnostic; the peptide exerts a spasmolytic effect and an inhibitory effect on gastric acid secretion similar to glucagon with not more than a negligible metabolic effect. The invention relates to a novel purified polypeptide which is recoverable from porcine pancreas glands. The amino acid composition of the said polypeptide has been determined to be as follows: Trp (2), Lys (4), His (1), Arg (5), Asx (10), Thr (3), Ser (9), Glx (12), Pro (12), Gly (6), Ala (6), Cys 1/2 (14), Val (7), Met (2), Ile (3), Leu (1), Try (2), Phe (7), wherein the determinations are subjected to the usual error of .+-.10 percent of the indicated figures. The partial amino acid sequence comprising a total of 45 amino acids from the N-terminal, is believed to be: ##STR1## wherein pyrGlu (residue 1) stands for pyroglutamic acid. The purified polypeptide is of utility as a medicament, for example as a spasmolytic agent, as an agent for treatment of gastroduodenal ulcers and as a diagnostic aid.
