Particles are released from a particle-containing area of a first surface of a porous matrix. The particle-containing area is contacted with a liquid medium and sonic energy is applied to an opposing area on a second surface of the porous matrix, wherein the opposing area is opposite to the particle-containing area. The particles may be biological particles or non-biological particles.

Particle release and collection

A reagent for detecting an analyte comprises a flavoprotein enzyme, a mediator such as a phenothiazine mediator, at least one surfactant, a polymer and a buffer. The reagent may be used with an electrochemical test sensor that includes a plurality of electrodes.

Reagents and methods for detecting analytes

A concentrated sample having enhanced concentration of the one or more different populations of target rare molecules is incubated with, for each different population of target rare molecules, a particulate or non-particulate affinity agent that comprises a specific binding partner that is specific for and binds to a target rare molecule. The affinity agent comprises a mass spectrometry (MS) label precursor or a first alteration agent, which either facilitates the formation of an MS label from the MS label precursor or releases an entity that comprises the MS label precursor from the affinity agent. A second alteration agent is employed if the first alteration agent does not facilitate the formation of an MS label from the MS label precursor. MS analysis is used to determine each different MS label.

Rare molecule detection

There is provided a system () and method () for improving ICC and/or ISH rare cell detection by lowering background noise and providing enhanced detection of rare cells (). In an embodiment, background noise for an ICC and/or an ISH rare cell detection assay is reduced and rare cell signaling is enhanced via feedback control. To accomplish the feedback control, an electronic control circuit () can direct a fluid delivery apparatus () to add an adjustment amount to account for fluid loss in the system () when fluid loss is indicated.

Feedback control for improved rare cell detection

A test strip or electrochemical sensor for measuring the amount of an analyte in a biological fluid, e.g. the glucose content of whole blood, includes a size self-limiting reagent formulation employing an enzyme system for reaction with the analyte, the reactive system mixed into a water-soluble swellable polymer matrix containing small water-insoluble particles having a nominal size of about 0.05 to 20 &#x3bc;m, preferably about 1 to 10 &#x3bc;m. The weight ratio of the water-insoluble particles to the water-soluble swellable polymer matrix is about 1/2 to 2/1. The reagent formulation is deposited onto a non-porous substrate to form a thin layer about 6-16 &#x3bc;m thick, providing a rapid and stable response to application of a sample, while being insensitive to the amount of the sample.

Size self-limiting compositions and test devices for measuring analytes in biological fluids

A method of forming a 3-phenylimino-3H-phenothiazine or a 3-phenylimino-3H-phenoxazine mediator includes providing a first reactant including phenothiazine or phenoxazine, providing a first solvent, providing a second reactant and providing a second solvent. The first reactant, first solvent, second reactant and second solvent are combined to form a reactants solution. Sodium persulfate is added to the reactants solution to couple the first and second reactants resulting in a reaction solution including the 3-phenylimino-3H-phenothiazine or the 3-phenylimino-3H-phenoxazine mediator.

Mediator for test sensor

A fluid sensor comprises a formed plastic body and a reagent. The body has a top face with an integral first surface. The body also has a bottom face opposed to the first surface and a sidewall that extends from the periphery of the top face. The first surface is adapted to accept a fluid sample. The reagent is disposed on the integral first surface and causes a color change detectable on the bottom face when the reagent reacts with an analyte in the fluid sample.

Glucose measured in blood samples is distinguished from glucose measured in the control solutions used to test the optical instruments which make such measurements. The control solutions contain a labeling substance recognized by the optical instrument to distinguish glucose measurements made of control solutions from those made of blood samples.
