The presently disclosed subject matter relates to systems for making an implantable construct comprising a reservoir coupled to the at least one frame; a mandrel base coupled to the at least one frame; a mandrel coupled to the mandrel base; a first mechanism configured to be coupled to the at least one frame and configured to move the at least one frame along a first axis; a second mechanism configured to be coupled to the at least one frame and configured to rotate the least one mandrel base along a second axis between a first orientation and a second orientation; and a container configured to be coupled to the at least one frame and configured to receive the mandrel in the second orientation. The disclosed mechanisms can manually, semi-automatically, or automatically control the movements of the at least one frame, reservoir, mandrel, mandrel base, and container to produce the construct with a lumen. The presently disclosed subject matter also relates to methods for making the construct.

Automation mechanism for pre/clinical production of resorbable nerve guides

Composite nerve guides for nerve regeneration are provided, wherein the composite guide comprise a nerve graft and a nerve conduit continuing an active agent that promote axon regeneration. The devices can provide structural supports to guide nerve regeneration and locally deliver an active agent (e.g., glial cell-line derived neurotrophic factor (GDNF) and/or glial growth factor 2 (GGF2) to injured nervous system tissue upon implantation in a subject. Methods of treatment using such devices are also provided.

Systems and methods for reconstruction of nerve defects

The inventions provided herein relate to compositions, methods, delivery devices and kits for repairing or augmenting a tissue in a subject. The compositions described herein are injectable such that they can be placed in a tissue to be treated with a minimally-invasive procedure (e.g., by injection) and/or be molded flexibly into a tissue void of any shape and/or size. In some embodiments, the composition described herein comprises a plurality of silk fibroin particles, which can retain their original volume within the tissue for a period of time. The compositions can be used as a filler to replace a tissue void, e.g., for tissue repair and/or augmentation, or as a scaffold to support tissue regeneration and/or reconstruction. In some embodiments, the compositions described herein can be used for soft tissue repair or augmentation.

The present invention provides a biocompatible material, comprising a phospholipid or phospholipid derivative comprising various functional groups having specific chemical properties, which can function as a modular surface design unit to be incorporated or appended to a desired substrate on which it is then polymerized, thereby contributing new or specified biochemical characteristics to the polymerized and stabilized biomaterial.

Blends of biodegradable polymers, preferably poly(caprolactone) and poly(D,L-lactic-co-glycolic) acid are discussed as well as their applications in the medical field, particularly with regard to bone tissue engineering. Preferably, hydroxyapatite ('HA') granules are incorporated into the blends and the resulting blends have desirable mechanical, physical, and biological characteristics. Even more preferably the compositions of the present invention are utilized to form osteoconductive composites that supported bone cell growth on the surface as well as throughout the scaffold.
