A method for autofocusing a microscope at a correct autofocus position in a sample includes the steps: generating a reference pattern by an autofocus light device, projecting the reference pattern towards a sample, whereby the reference pattern is backscattered by at least two interfaces being located at or close to the sample, projecting the backscattered reference pattern towards a detector which provides spatial resolution, obtaining a superposition of a number of detection patterns, each detection pattern related to one of the interfaces, on the detector, analyzing the superposition of detection patterns to identify at least one autofocus detection pattern related to at least one of the interfaces, and analyzing the at least one autofocus detection pattern to determine the direction and/or magnitude of deviation of the microscope's current focus position from the correct focus position.

Method for autofocusing a microscope at a correct autofocus position in a sample

A device for measuring the optical properties of an object, particularly of a chemical and/or biological sample, comprises an illumination light source for illuminating the object. Further, an excitation light source is provided for illuminating the object, wherein the radiation of the excitation light source being suited to change the optical properties of the object. Further, the excitation radiation is focused in a measurement volume arranged within the object. By means of a detector device, there can be detected preferably the transmission of the illumination radiation through the object within the measurement volume.

Device and method for measuring the optical properties of an object

An optical measurement device for measurement of chemical and/or biological samples comprises an illumination means () for illuminating the sample () to be measured. Further, an optics means () is provided for imaging a sample area in an image field (). A first partial area () of the image field is covered by a first detector (), and a second partial area () of the image field () is covered by a second detector (). Thus, it is possible to change between two measurement methods without switching or to perform two measurements on a sample at the same time.

Device and method for optical measurement of chemical and/or biological samples

A confocal microscope has a specimen holding device for holding a specimen (). The specimen () is illuminated by an illuminating unit (). An optics unit () serves to direct radiation produced by the illuminating unit () toward the specimen () and to direct the radiation emitted by the specimen toward a detector unit (). The confocal microscope also comprises an aperture diaphragm () that is placed in the beam path in front of the detector unit (). In addition, a focusing lens () is provided in the beam path in front of the aperture diaphragm (). The focusing lens () can be moved in order to adjust the confocal microscope, for example, in order to compensate for thermal stresses.

In a method for analyzing a chemical and/or biological sample, particularly in high- and medium throughput screening systems, an observation beam () is focused in an observation volume () of the sample. For analyzing the sample, the observation beam () is moved in the sample. According to the inventions, to improve the quality of the measurement, the observation beam is moved continuously in the observation volume ().
