DNA2 inhibitors for cancer treatment

Inhibition of DNA2 in Fanconi anemia (FA) cells remedies the over-resection of DNA, thereby stabilizing the FA cells. Inhibition of DNA2 in FA cells allows for safe treatment of cancers in FA patients, a decrease in the lethality of FA cells, a decrease in bone marrow failure of FA patients, and a means for decreasing the incidence of cancer for FA patients.

Inhibition of DNA2 in fanconi anemia

The invention provides novel DNA polymerases of the &bgr;-family, called &bgr;-type DNA polymerases. Nucleic acids encoding &bgr;-type DNA polymerases are provided, as well as expression vectors and host cells containing the nucleic acid encoding the &bgr;-type DNA polymerases. The invention further relates to methods for synthesizing nucleic acid using &bgr;-type DNA polymerases.

&bgr;-type DNA polymerases

Compositions for enhancing the production of toxic polypeptides in prokaryotic hosts comprise a coding nucleic acid and a bacteriophage nucleic acid. The coding nucleic acid encodes a promoter operably linked to a gene encoding a selected toxic protein. The bacteriophage nucleic acid contains at least one gene from the early region of a T7-like bacteriophage other than an RNA polymerase. When both the coding nucleic acid and the bacteriophage nucleic acid are used to transform or transfect a prokaryotic host, the production of the toxic polypeptide is greater than the level of production of the same toxic polypeptide when the same host is transformed only with the coding nucleic acid.
