Systems, apparatuses, kits, and methods for purification and analysis of analytes having a broad range of hydrophobicities by liquid chromatography-mass spectrometry (LC-MS). Using one set of liquid chromatography columns, one set of mobile phase buffers, and, optionally, a single ionization method (e.g., electrospray ionization), a wide range of analytes can be purified and analyzed on a liquid chromatography-mass spectrometry (LC-MS) system. LC-MS purification and analysis of analytes having a broad range of partition coefficients is accomplished by selecting LC run parameters and MS system parameters that are particular to different classes of analytes without having to make column or buffer changes or any other hardware configuration changes to the LC-MS system. The methods, systems, and kits described herein provide for substantially increased speed/throughput and ease of use for a wide range analytes with essentially no compromise in specificity for individual analytes relative to previously described methods.

LC-MS configuration for purification and detection of analytes having a broad range of hydrophobicities

A method for analyzing and quantifying a panel of drugs in a clinical sample comprises: trapping a first portion of drug parent compounds and their metabolites on a first chromatographic column; trapping a second portion of the drug parent compounds and their metabolites on a second chromatographic column; separately eluting the first and second portions of the drug parent compounds and their metabolites from the first and second chromatographic columns; detecting concentrations of each of the drug parent compounds and metabolites eluted from each of the first and second chromatographic columns with a detector; and summing the detected concentration of each drug parent compound together with the detected concentrations of all of its respective analytes so as to derive a respective total concentration of each drug in the sample.

Methods for analysis of phase-I and phase-II metabolites and parent compounds without hydrolysis

A method of multiplexing the operation of a sample preparation and analysis system is disclosed. The system includes a sample preparation system capable of preparing a first sample in accordance with a first assay that is selected from a database containing a plurality of unique assays and preparing a second sample in accordance with a second, different assay. The method includes separately transporting the first and second prepared samples to an analysis station having first and second separation channels and an analyzer comprising a mass spectrometer. The method further includes simultaneously analyzing the first prepared sample with the analyzer in accordance with the first selected assay and separating the second prepared sample with the second separation channel in accordance with the second selected assay.

An apparatus comprises: a first chromatographic column fluidically coupled to a source of sample and a source of a first chromatographic mobile phase solvent; a second chromatographic column fluidically coupled to the first column; a source of a second mobile phase solvent fluidically coupled between the first and second columns; and a detector, the first chromatographic column being configurable to receive, in a trapping step, the first solvent and sample and to retain a first portion of a plurality of analytes therein and to pass a second portion of the plurality of analytes therethrough, the second chromatographic column being configurable, in the trapping step, to receive the second portion of the plurality of analytes and the first and second solvents and to retain the second portion of the plurality of analytes therein, the detector being arranged to receive the second and first portions in respective first and second elution steps.

System for analysis of phase-I and phase-II metabolites and parent compounds without hydrolysis

Systems, kits, and methods for quantitation of metabolites of vitamin D by liquid chromatography-mass spectrometry (LC-MS). The systems, kits, and methods described herein stabilize and/or promote the formation of the protonated molecular ion ([M+H]+) for the vitamin D metabolites in the ionization source (e.g., electrospray ionization ('ESI')). Formation of the molecular ion does not involve loss of a water molecule from the parent molecule. Subsequent fragmentation of the [M+H]+ ion yields product ions that are specific to each molecular ion. The systems, kits, and methods described herein provide for no compromise in specificity and provide for a significant increase in sensitivity relative to previously described methods.

LC-MS separation and detection of vitamin D metabolites

A non-aqueous, particle-forming, fused pyrrolocarbazole-containing composition is disclosed. Upon contact with an aqueous medium, the particle-forming composition spontaneously disperses into suspended particles, thereby forming a stable suspension that provides greatly improved bioavailability of orally administered fused pyrrolocarbazole compounds.

Disclosed herein are aqueous indolocarbazole solutions. In one embodiment, the invention features a solution comprising: (1) an indolocarbazole; (ii) a selected organic solvent being present in a concentration of between about 1% and about 99% by weight inclusive, (iii) a dispersant being present in a concentration of between about 0.25% and about 10% by weight inclusive; (iv) water being present in a concentration of between 0% and about 99% by weight inclusive, and (v) a polyethylene glycol being present in a concentration of between 0% and about 60% by weight inclusive.
