Pro-fibrotic coatings for medical articles are described that include a pro-fibrotic polymer such as collagen. The pro-fibrotic coatings can also include a thromboresistant polymer. The coatings can be formed by activation of photoreactive groups pendent or independent of the coating materials.

The invention provides methods and compositions for providing biocompatible surfaces to medical articles. In particular the invention provides biocompatible coatings with heparin activity. In some aspects, the biocompatible coatings of the invention are able to release a bioactive agent. The invention also provides methods for improving the quality of bioactive agent-containing coatings by performing pre-irradiation of biocompatible coating compositions.

Process and systems for biocompatible surfaces

A crosslinkable macromer system and related methods of preparing the system and using the system in the form of a crosslinked matrix between a tissue site and an implant article such as a tissue implant or on the porous surface of a prosthetic device. The macromer system includes two or more polymer-pendent polymerizable groups and one or more multifunctional initiator groups. The polymerizable groups and the initiator group(s), when polymer-pendent, can be pendent on the same or different polymeric backbones. The macromer system provides advantages over the use of polymerizable macromers and separate, low molecular weight initiators, including advantages with respect to such properties as nontoxicity, efficiency, and solubility. A macromer system of the invention can be used as an interface between the tissue site and implant article in a manner sufficient to permit tissue growth through the crosslinked matrix and between the tissue site and implant. In a preferred embodiment, polymers with pendent polymerizable groups, for use in the macromer system, are prepared by reacting a polysaccharide polymer with a reactive moiety in an organic, polar solvent such as formamide.

Pannus-resisting implantable medical devices comprise one or more antimicrobial reservoirs, each such reservoir incorporating antimicrobial substances in predetermined distributions for timed release in vivo. Predetermined distributions of antimicrobial substances incorporated in antimicrobial reservoirs are achieved through use of fluid solvent carriers, which may comprise supercritical fluid solvents. Precipitation of antimicrobial substances from such solvent carriers in predetermined distributions is accomplished through evaporation and/or application of heating, cooling or decreased ambient pressure to the solvent carriers.

A prosthetic heart valve having an elastomeric stent with an annular base and a plurality of apertures or holes circumferentially spaced around the base for receiving a suture. Flexible polymeric leaflets are molded over the stent. Polymeric material surrounds the apertures, leaving a central opening in each aperture. The base may be a frustro-conical ring. A sewing ring is mounted on the heart valve by suturing the ring through the apertures. Apertures or holes may also be provided by extending an area covered by woven material around all or part of the circumference of the base.

A biocompatible heart valve is described having incorporated therein an effective amount of applied coating to render the valve resistant to in vivo pathologic thrombus formation and resistant to in vivo pathologic calcification.

Biocompatible prostheses, specifically, biocompatible heart valves, are described which comprise a blend of a polyurethane (PU) and a polysiliconeurethane (PSU). Preferably, the polyurethane is a polyetherurethane (PEU). More preferably, it is a polyetherurethane urea (PEUU). Such prostheses typically exhibit reduced mineralization or thrombosis and/or reduced biological degradation.

Polymeric heart valve fabricated from polyurethane/polysiliconeurethane blends

A method is provided for producing antimicrobial, antithrombogenic medical devices. The method employs an antimicrobial treatment process wherein an antimicrobial agent is dissolved in an appropriate solvent and the resulting solution is contacted with a portion of a medical device of interest. The antimicrobial treatment process is advantageously performed without the need for additional compounds to facilitate antimicrobial agent uptake into the device. The method further comprises an antithrombogenic treatment process wherein antithrombogenic agents or materials are applied or otherwise associated with at least some portion of the medical device.

Method for producing antimicrobial antithrombogenic medical devices

This invention provides an antimicrobial annuloplasty rings, and methods for making the same, wherein the annuloplasty rings have a desired degree of initial rigidity to facilitate ease of handling during implantation but which becomes flexible some time after implantation. The annuloplasty ring contains a relatively rigid insert enclosed by a fabric sheath, the insert being at least partly comprised of a biodegradable material. Following surgical implantation of the annuloplasty ring, the rigid insert component of the ring, upon exposure to blood and/or other physiological fluids, undergoes a controlled biodegradation which decreases its rigidity, thereby increasing the flexibility of the implanted annuloplasty ring. Furthermore, at least some portion of the annuloplasty ring of the invention has incorporated therein one or more antimicrobial agents in a manner which reduces the likelihood of device infection following implantation.
