The invention provides a structured cell growth matrix or assembly comprising a one or more spacer layers and one or more cell immobilization layers. The invention further provides a bioreactor comprising said matrix or assembly.

Cell growth matrix

A sampler for use with a bioreactor for growing a cell culture is disclosed. In one embodiment, the bioreactor includes a structured fixed bed including a removable sample portion for recovering a sample of cells from the cell culture. Related apparatuses and methods are also disclosed.

Fixed bed sampler and related methods

An apparatus for culturing cells includes a bioreactor. The bioreactor may be modular and may include in a chamber a fixed bed, such as an unstructured or structured fixed bed (such as a spiral bed) for culturing cells, with a return column arranged centrally within the chamber. The modular bioreactor may include a plurality of structured fixed bed arranged in a stacked configuration. The modular bioreactor may include an outer casing forming a space for conditioning (e.g., insulating, heating, cooling) at least a chamber in which cells are cultured. The bioreactor may also include an impeller with radially curved blades, and may also suspend the impeller so that it may move from side-to-side and align with an external drive. Related methods are also disclosed.

Laboratory instrument

A system for clarifying a cell culture harvest solution, including target molecules and dynamic filter media. A filtration vessel includes a flexible liner and at least one filter having a surface on which the dynamic filter media accumulates into a cake. The cake and filter during filtration operation permit a filtrate including target molecules to pass therethrough while preventing unwanted solid materials from passing therethrough. A backflush source includes a backflush fluid and is fluidly connected to the filtration vessel via the at least one filter. The backflush source, during backflush operation, is adapted to supply backflush fluid back through the at least one filter for removing the cake formed on the filter. Related systems and methods are also disclosed.

System and method for clarifying a cell culture harvest solution

A method and a kit for cell culture clarification, as well as a system or apparatus for clarifying a cell culture and purifying biomolecules of interest, are described. The cell culture comprises at least one biomolecule of interest. The method comprises adding one or more compounds to the cell culture that allow the formation of floccules of solutes and/or particulates in said cell culture harvest, adding an amount of diatomaceous earth (DE) to said cell culture harvest, agitating the obtained solution, and transferring said solution to a filtration vessel comprising a support filter having a surface, whereby the diatomaceous earth in said solution forms a layer on said surface and filtering said harvest solution through said layer and support filter.

Cell culture clarification

The current disclosure concerns a system for producing biomolecules comprising a bioreactor including a chamber () suitable for receiving a liquid comprising cells and viral particles; and a concentrator (), wherein said concentrator is equipped with a retentate conduit () suitable for collecting said retentate and facilitating recirculating of the retentate to an input of said bioreactor or to an input of an intermediate vessel () positioned between said concentrator and said bioreactor. In a second and third aspect the disclosure concerns a method for producing biomolecules and the use of the disclosed system for the production of biomolecules.

System and method for the production of biomolecules such as viral vaccines

The invention provides a method and a kit for combined virus inactivation and capture/purification of a feed containing a protein of interest. The method comprises the steps of: contacting a feed comprising a protein of interest with negatively charged particles or media, conditioning the particles or media such that the pH is acidic thereby inactivating said virus, and eluting said protein of interest.

Protein purification method and kit

The invention provides a method and a system for the production of viruses or virus derived products. The system comprises a containment enclosure provided with a single entry means and a single exit means which are movable between a closed position and an open position. The containment enclosure comprises at least one production unit comprising at least one bioreactor for culturing the virus, said bioreactor is provided with at least one inlet and at least one outlet; at least one purification unit fluidly connected to the production unit and comprising at least one purification means having at least one inlet and at least one outlet. The system further comprises at least one inactivation and/or at least one gas decontamination unit. The containment enclosure comprises at least one process control device connected to the exit means. The movement of the exit means from the closed position to the open position is controllable by said process control device.

System for the production of cells and/or cell products

A disposable material processing apparatus, useable as a bioreactor or fermenter, includes a hollow tank () and a mixing paddle () disposed within the interior of the tank and adapted to mix contents therein. The paddle may be isolated within a flexible sleeve (). Various functional elements, such as a sparger, sensor, material extraction conduit, material addition conduit, and/or heat exchange element may be provided, and optionally arranged to travel with the paddle within the tank interior. Baffles may protrude into a mixing tank to enhance mixing. A tank and/or sleeve may comprise polymeric film materials.
