The present invention reveals human recombinant antibodies that recognize the human Vascular Endothelium Growth Factor A (VEGF-A), block its interaction with the VEGFR2 receptor, and interfere with its proliferative effect in vitro and pro-angiogenic effect in vivo. The antibodies identify an epitope on human VEGF-A different from any other previously reported, and were obtained by combining one immunoglobulin light chain variable region with other three heavy chain ones. The antibodies were obtained by human immunoglobulin variable region mutagenesis, and can be employed for the immunotherapy of pathological entities associated with an increase in vasculature, such as age-related macular degeneration, cancer, and others.

Recombinant antibodies to the vascular endothelial growth factor (VEGF) which are obtained by means of mutagenesis of variable regions

Application of oligonucleotide and polypeptide sequences of molecules of the family of the vascular permeability factor (VPF), their receptors, and co-receptors, as well as their modifications, in the active immunotherapy of pathologic entities in which course is associated to the increase of angiogenesis. These procedures can be employed in the single or combined therapy for the treatment of cancer and its metastasis, acute and chronic inflammatory processes, infectious diseases, autoimmune diseases, diabetic and newborn retinopathies, organ transplant rejection, macular degeneration, neovascular glaucoma, hemangioma, and angiofibroma, among others.

Active antiangiogenic therapy

The present invention is related to a composition capable of inhibiting the growth of tumoral cells of different histological origins and of activated endothelial cells. The components of said compositions are polypeptide fragments of the serralisins, corresponding to the C-terminal fragment, from the internal metionine trough the end of the molecule, which could be combined among them and optionally with the prodigiosins that potentiate the antitumoral effect of the composition. The prodigiosins in the composition could be at a concentration of 0.1-100 nM. The anti-proliferative action of this composition is mediated by apoptotic mechanism. It's 'in vivo' administration has antitumoral, antiangiogenic and protective effect against malignant tumors.

The recombinant antibodies were obtained using recombinant DNA technology and are characterized in that they recognize antigen ior C2. The recombinant antibodies are specifically chimeric antibody, humanized antibody, and single chain Fv fragment. The chimeric antibody contains the variable domains of the murine immunoglobuline and the constant regions of the human immunoglobuline. The humanized antibody contains the constant regions of human immunoglobuline and has been specifically modified in the murine frameworks regions (FRs) and within the latter, in those areas that may result in an antigenic site for cells T. The Fv fragment contains the variable domains of murine immunoglobuline. The invention also relates to the utilization of recombinant antibodies derived from murine antibody ior C5 in the diagnosis and therapy of colorectal tumors, the metastasis thereof and recurrences.
