A phase plate for high precision wavelength extraction can include a planar substrate which has a point spread function engineered profile formed of a tessellation of regions. The point spread function engineered profile transforms a point spread function of a light source to form a wavelength dependent geometric pattern. The geometric pattern can also preserve spatial location information of the light source. Such a phase plate permits extracting three-dimensional position and the wavelength of a point emitter.

A microscopy system which includes a light source for illuminating a sample; an objective lens for capturing light emitted from the illuminated sample to form a signal beam; and a dispersive optical element through which the signal beam is directed, wherein the dispersive optical element converts the signal beam to a spatially coherent signal beam.

Coherent fluorescence super-resolution microscopy

Methods and systems for improving high resolution imaging using a polarization-modulated tip enhanced optical microscope. A polarizer is configured to alternately create and remove a region of enhanced optical intensity adjacent the tip of the microscope probe at the focus of a light source. The sample being studied emits photons at specific rates relative to a background rate depending on the existence or nonexistence of the region of enhanced optical intensity. Comparing the rate of emissions when the region of enhanced optical intensity exists to when it does not creates a detailed image of the sample. By not requiring the probe to oscillate, this system enhances the resolution of the microscope without potentially causing damage to the sample.

Polarization-modulated tip enhanced optical microscope

The conversion of linear polarized light into radial-polarized light is accomplished through a radial polarization generator containing multiple layers of polarizing film, or polarization converters, for converting the beam of light to a different orientation. Using the radial polarization generator, a quasi radial-polarized beam of light can be obtained quickly and easily by simply aligning the light at the center of the polarization generator. True radial-polarization can be obtained by passing the laser through a spatial filter. With such a radial polarization generator, a large frequency range of light may be converted from a linear orientation to a radial orientation.
