Apparatuses, systems, and methods for an open-top light-sheet (OTLS) microscope which includes an illumination objective and a collection objective which have optical axes which are non-orthogonal to each other. The optical axis of the collection objective may be orthogonal to a plane of the sample holder. The illumination and collection objective may be located below the sample holder. The OTLS microscope may optionally include a second collection objective which has an optical axis orthogonal to the optical axis of the illumination objective. The illumination objective may be an air objective, and the collection objective may be an immersion objective.

Open-top light-sheet microscopy with a non-orthogonal arrangement of illumination and collection objectives

Apparatuses, systems, and methods for solid immersion meniscus lenses (SIMlenses). An optical system may include a sample holder with a first side which supports a sample, and a second side opposite the first side. The second side of the sample holder may be in contact with an immersion fluid. Light passing between the sample and an objective lens may pass through the sample holder, immersion fluid, and a SIMlens positioned between the immersion fluid and objective. The SIMlens may have a first curved surface and a second curved surface, each of which may be shaped to match a wavefront of the light as it passes through the SIMlens. The immersion fluid, SIMlens, and environment containing the objective may all have different refractive indices.

Apparatuses, systems and methods for solid immersion meniscus lenses

Embodiments disclosed herein directed to multi-direction digital light sheet microscopy (mDSLM). In mDSLM a sample may be illuminated with an illumination sheet which has a first angular diversity along a first axis and a second angular diversity along a second axis which is orthogonal to the first axis. An mDSLM system includes beam shaping optics which may passively reshape light into the illumination sheet. In some embodiments, scanning optics may be used to scan the illumination sheet with respect to the sample. In some embodiments, a confocal aperture may be used to limit out of focus light which reaches a detector of the mDSLM system.

Apparatuses and methods for multi-direction digital scanned light sheet microscopy

Apparatuses, systems, and methods for dynamically shaped focal surface with a scanning microscope. A microscope (e.g., a scanning confocal microscope) may use a scanning element to scan an illumination beam to generate a focal region. The scanning element may include multiple degrees of freedom, such as rotation and translation along orthogonal axes. For example, if the illumination beam is a line focus, rotation along a first axis and translation along a second orthogonal axis may sweep the line focus across a focal surface which is substantially flat and normal to the second axis. In some embodiments, the microscope may be a dual-axis handheld microscope, where a single objective in a handheld housing is used to both direct the scanned illumination beam and receive the collected light.
