The present invention relates to methods for controlling hybridization in plants and producing hybrid plants. The present invention also relates to nucleic acids encoding amino acid sequences for self-incompatibility (SI) proteins in plants, and the use thereof for the manipulation of SI, including seed production, in plants, particularly of the Poaceae family. The present invention also relates to kits, compositions, constructs and vectors including such nucleic acids, and related polypeptides, regulatory elements and methods. The present invention also relates to expression of self-gamete recognition genes in plants and to related nucleic acids, constructs, molecular markers and methods.

Manipulation of self-incompatibility in plants

A method of gene editing or gene stacking within a FAD3 loci by cleaving, in a site directed manner, a location in a FAD3 gene in a cell, to generate a break in the FAD3 gene and then ligating into the break a nucleic acid molecule associated with one or more traits of interest is disclosed.

The present invention relates to a method for identifying and/or characterising an endophyte strain, said method including providing a plurality of samples of endophytes, subjecting said endophytes to genetic analysis; subjecting said endophytes to metabolic analysis and selecting endophytes having a desired genetic and metabolic profile. The present invention also relates to novel endophytes having a desired toxin profile wherein the endophyte produces significantly less toxic alkaloids compared with a control endophyte such as standard toxic (ST) endophyte; and/or significantly more alkaloids conferring beneficial properties compared with a control endophyte such as ST endophyte. The present invention also relates to endophyte variants having a desired genetic and metabolic profile, wherein said endophyte variants possess genetic and/or metabolic characteristics that result in a beneficial phenotype in a plant harbouring or otherwise associated with the endophyte variant. Preferably said endophyte variants are generated by polyploidisation or induced chromosome doubling.

Endophytes and related methods

An Engineered Transgene Integration Platform (ETIP) is described that can be inserted randomly or at targeted locations in plant genomes to facilitate rapid selection and detection of a GOI that is perfectly targeted (both the 5' and 3&#x2032; ends) at the ETIP genomic location. One element in the subject disclosure is the introduction of specific double stranded breaks within the ETIP. In some embodiments, an ETIP is described using zinc finger nuclease binding sites, but may utilize other targeting technologies such as meganucleases, CRISPRs, TALs, or leucine zippers. Also described are compositions of, and methods for producing, transgenic plants wherein the donor or payload DNA expresses one or more products of an exogenous nucleic acid sequence (e.g. protein or RNA) that has been stably-integrated into an ETIP in a plant cell. In embodiments, the ETIP facilitates testing of gene candidates and plant expression vectors from ideation through Development phases.

Engineered transgene integration platform (ETIP) for gene targeting and trait stacking

The present invention relates to new methods of selecting and breeding organisms, in particular organisms which exhibit symbiotic behaviour with symbionts, and to new organisms and symbiota developed thereby, such as plant or grass and endophyte symbiota. Multiple symbionts are deployed in multiple organisms and selected for improved symbiotic compatibility and performance early in the breeding process. Methods include producing improved organisms from germplasm, by inoculating host organism germplasm libraries with symbionts selected from symbiont libraries and selecting improved host organisms exhibiting desired symbiota characteristics, and selection of organism-symbiont associations with a desired genetic and metabolic profile by metagenomic analysis of nucleic acid libraries from an organism or organism-symbiont association.
