A system, methods, and apparatus are described to collect and prepare single cells, nuclei, subcellular components, and biomolecules from specimens including tissues and in some embodiments use the single cells to form organoids or microtissues. The system can perform enzymatic and/or physical disruption of the tissue to dissociate it into single-cells and then use a hanging droplet method to form organoids or microtissues.

Method and apparatus for processing tissue samples

An apparatus and method for performing successive chemical or biochemical operations in an automated or semi-automated format are disclosed. The apparatus includes (i) a substrate having two or more wells, where each well is separated from adjacent well(s) by an energy barrier, (ii), a magnetic bead having a surface on which at least one of said operations is performed, and (iii) electromagnets positioned adjacent said wells, for use in producing controlled-time electromagnetic fields in one or more selected wells. A control unit operatively coupled to the electromagnets functions to produce controlled-time electromagnetic forces on the bead that are effective to transfer the bead from one well to another across the energy barrier and optionally, to agitate the bead within a well.

Device and method for handling reaction components

The invention relates to microfluidic devices and methods that employ electromagnetic radiation to move a droplet of fluid on a fluid-transporting surface of a substrate. Typically, radiation of a particular wavelength is directed through a substantially transparent material, and the radiation imparts an optical trapping force to move the droplet. In addition, a means for reducing evaporative loss from the droplet may be provided.

The apparatus and method of the present invention disclose a system in which multiple injections may be made into a capillary array. The injections are spaced in time with each injection followed by an interval of electrophoresis. Once all samples are loaded into the capillaries, continuous electrophoresis and detection is used to separate and detect target compounds within the sample. The interval between injections is matched to the target compound migration rate to be sufficient to allow the target compounds to be detectably separated when the compounds reach the detector.

A capillary electrophoresis separation matrix for single-stranded nucleic acids, along with methods for using and preparing the matrix, are disclosed. The separation matrix provides denaturing conditions and contains hydroxyethyl cellulose (HEC) in combination with urea, and preferably also includes formamide. The separation matrix may be used for DNA sizing and sequencing applications and provides a single-base resolution to approximately 500 base pairs. The separation matrix is inexpensive, easy to prepare, requires no polymerization steps, and is of low enough viscosity to be pumped easily into and out of capillary tubes for electrophoresis. The low viscosity allows for high throughput of samples and reuse of the capillary tubes for numerous separations.
