The present invention is in the field of genetics, especially plant genetics, and provides agents capable of controlling gene expression. The present invention specifically provides sequences of naturally occurring, tissue-specifically expressed microRNAs. The invention further provides for transgenic expression constructs comprising sequences encoding such microRNAs. By incorporation of the microRNA encoding sequence the expression from the expression construct is specifically silenced in the tissue where the naturally occurring microRNA is naturally expressed. Thereby the expression profile resulting from the promoter is modulated and leakiness is reduced. The invention further provides for a method for modulating transgenic expression by incorporating sequences encoding the microRNAs into transgenic expression constructs. The compositions and methods of the invention can be used to enhance performance of agricultural relevant crops and for therapy, prophylaxis, research and diagnostics in diseases and disorders, which afflict mammalian species.

Methods controlling gene expression

The present invention is in the field of genetics, especially plant genetics, and provides agents capable of controlling gene expression. More specifically the inventions relates to methods for engineering ta-siRNA primary transcripts in order to target gene-of-interest (GOI) and control their expression. The invention further provides for a method for modulating transgenic expression by said engineered ta-siRNAs.

Methods for controlling gene expression using ta-siRNA

The invention relates to efficient, high-throughput methods, systems, and DNA constructs for identification and isolation of transcription termination sequences. The invention relates further to specific terminator sequences identified by said methods isolated from rice.

A release mechanism that decreases the effort necessary to release a tensile load is provided. The mechanism includes a connecting body that secures a member connected to a load between a fixed jaw and an opposing jaw, two floating links connected to a moveable jaw and to a moveable toggle pivot, and a moveable toggle that includes a central lever and two toggle links mounted on the moveable toggle pivot. A cam lobe is formed on the lever and rotation of the cam lobe against a flat rise included in the connecting body decreases the effort necessary to unlock the mechanism and release the load. The central lever is interspersed between the toggle links and can move between spacer/stops attached to each of the toggle links. The spacer/stops keep the lever from exceeding the prescribed range of motion.

Specifically, the invention relates to the use of RNA interference to inhibit expression of a target essential parasitic nematode gene, which is a parasitic nematode pas-5 gene, and relates to the generation of plants that have increased resistance to parasitic nematodes.

Compositions and methods using RNA interference for control of nematodes

The present invention concerns double stranded RNA compositions and transgenic plants capable of inhibiting expression of essential genes in parasitic nematodes, and methods associated therewith. Specifically, the invention relates to the use of RNA interference to inhibit expression of a target essential parasitic nematode gene selected from the group consisting of a parasitic nematode cytosolic chaperonin gene, a parasitic nematode gene encoding heat shock protein-90, a parasitic nematode gene homologous to theY65B4BR.5a gene, and a parasitic nematode gene homologous to apat-10 gene, and relates to the generation of plants that have increased tolerance to parasitic nematodes.

An improved toggle locking release mechanism (8) is provided, including a connecting body (10), and opposing pivoted jaw (22), and multi-pivoted floating links or movable lock/release links (30) and a movable toggle link/release lever (34). The mechanism of the present invention is adapted to provide a secure locking connection as well as facilitating multi-directional quick releasing while the mechanism (8) is under high-tensile load.

A three-part light switch cover for enabling remote operation of a wall mounted toggle-type electrical switch comprising an elongated wall plate member adapted to enclose safely a sliding member that includes an upper end portion having an aperture adapted to receive a light switch lever and a lower end portion adapted so that the sliding member, formed at the lower end with an operating handle, is restricted to and accessible through the wall plate member. The wall plate member and sliding member are extended downward from an existing switch apparatus allowing a child or disabled person to operate the light switch from a height beneath the switch. Full use of the light switch is retained by the present invention for utilization in standard manner by means of an enlarged aperture in the wall plate member receiving the light switch lever. The wall plate member and sliding member are easily installable on a permanent, as well as temporary, basis.

An improved releasable toggle locking mooring hook (10) having a releasable hook (20), toggle linkage (89), a stop/release mechanism (119), a self locking safety latch (126) and a self release mechanism (251) adapted to provide a secure locking mooring hook and a low friction, low effort releasing means while mooring hook is under heavy tensile load. In the mooring of ships, boats and barges to a dock or oil well platform, a line, usually under tension, is run from the ship to a mooring means mounted on the dock or platform. When it is desired that the ship leave this moorage, this mooring means can be actuated manually, mechanically or by a pre-set tensioned self releasing means to release mooring line and thus, allow the ship to sail. This mooring means is identified as a releasable toggle locking mooring hook (10) of this invention.
