Medical devices, substrates and biologic therapies for bone repair and guided tissue regeneration are disclosed. More particularly, bone graft substitutes and bone void fillers which comprise a porous collagen matrix and calcium deficient hydroxyapatite ceramic granules for delivery of osteoinductive or other therapeutic agents are disclosed.

Systems and methods for preparing synthetic osteoinductive bone grafts are provided in which a porous ceramic granule, which may be incorporated within a biocompatible matrix material, is loaded with an osteoinductive agent. Loading of granules is facilitated in some cases by the use of low-pH buffers and pre-treatments.

Systems and methods for improved delivery of osteoinductive molecules in bone repair

The invention relates to novel designer osteogenic proteins having altered affinity for a cognate receptor, nucleic acids encoding the same, and methods of use therefor. More preferably, the novel designer osteogenic proteins are designer BMPs and have altered affinity for a cognate BMP receptor. The designer BMPs demonstrate altered biological characteristics and provide potential useful novel therapeutics.

Osteoconductive synthetic bone grafts are provided in which porous metallic matrices are loaded with an osteoinductive protein. In certain embodiments, the grafts include porous ceramic granules deposited within the matrices.

Osteogenic proteins

An injectable formulation is disclosed for delivery of osteogenic proteins. The formulation comprises a pharmaceutically acceptable admixture of an osteogenic protein; and formulations comprising osteogenic protein, hyaluronic acid derivatives and tricalcium phosphate are also disclosed. Methods for formulating porous injectable gels and pastes from hyaluronic acid are also disclosed.

Purified cartilage and/or bone inductive proteins and processes for producing them are disclosed. The proteins may be used in the treatment of bone and/or cartilage defects and in wound healing and related tissue repair.

Compositions of proteins with tendon/ligament-like tissue inducing activity are disclosed. The compositions are useful in the treatment of tendinitis and tendon or ligament defects and in related tissue repair.

Purified BMP-2 and BMP-4 proteins and processes for producing them are disclosed. The proteins may be used in the treatment of bone and cartilage defects and in wound healing and related tissue repair.
