The present relates to an autologous bioadhesive sealant composition, or fibrin glue prepared by a two-phase method, wherein all of the blood components for the bioadhesive sealant are derived from a patient to whom the bioadhesive sealant will be applied. A platelet rich plasma and a platelet poor plasma are formed by centrifuging a quantity of anticoagulated whole blood that was previously drawn from the patient. In one embodiment the platelet rich plasma is divided into two portions. In phase one, a compound that reverses the effect of the anticoagulant is added to the first portion and a clot is allowed to form. The clot is then triturated, and the resulting, serum containing autologous thrombin is collected. In phase two, the serum obtained from phase one is mixed with the second portion of the-platelet rich plasma to form the bioadhesive sealant of the invention.

Methods of applying a biological composition to an individual

The delivery system of the present invention is comprised of at least two chambers which store, individually, an activated blood component and an inactivated blood component. The first chamber comprises an activating agent and a filter which may be one in the same thus allowing the formed clot to be triturated thereby isolating the thrombin from the clot. The second chamber stores the inactivated blood component that when mixed with thrombin will produce a gel. The first or second chamber may further contain agents which are desired to be delivered to a specific site. The unique design of the delivery system allows for ease in operation of combining two agents at a specific time and place.

Autologous platelet gel spray delivery system

Provided are methods of applying biological compositions, that is, autologous bioadhesive sealant compositions containing one or more biological agents, to an individual, wherein all the blood components used in preparing the composition are derived from the patient who is to receive the biological composition. In one embodiment, the method comprises obtaining a whole blood sample from an individual; forming an inactive platelet rich plasma from the whole blood sample; mixing a biological agent into the inactive platelet rich plasma; obtaining thrombin from the whole blood sample; mixing the thrombin into the inactive platelet rich plasma to form a biological composition; and applying the biological composition to the individual.

Autologous fibrin sealant and method for making the same

A system for the production of a blood component composition is provided. The system includes a centrifuge having a blood reservoir for receiving and separating a blood sample into multiple components; a dispenser disposed outside of the centrifuge having a first collection chamber containing an activation agent and a second collection chamber containing one or more medicinal agents; means for removing a first portion at least one separated blood component from the centrifuge to the first container and a second portion to the second collection chamber, wherein the first collection chamber activates the first portion and stores the resulting clot and thrombin; a filter for separating the thrombin from the clot; and a nozzle for entraining and mixing the thrombin with the second portion containing the one or more medicinal components.

A blood separation system is fully mechanized to collect blood from a patient, separate waste portions of the blood, wash the blood, and redirect the usable portions to a device for reinjecting the usable portions into the patient. The system provides screen displays with detailed setup instructions and instructs the operator at the appropriate times to do certain manual steps. Apparatus for sequestration of platelet rich plasma spins at a high speed sufficient to separate solid cells from the blood sample and then spins at a lower speed for a predetermined time to allow platelets to elute from the solid cells.

A system for the production of autologous thrombin, comprising a centrifuge including a blood reservoir for receiving and separating an autologous anticoagulated blood sample having multiple inactive blood components and means for removing at least one of said inactive blood components upon separation, and a dispenser having at least two collection chambers for receiving said at least one of said inactive blood components, wherein a first collection chamber activates a first portion of said inactive blood component and stores the resulting coagulated blood component comprising a clot and said autologous thrombin.

First a platelet rich plasma and a platelet poor plasma are formed by centrifuging a quantity of anticoagulated whole blood that was previously drawn from the patient. The platelet rich plasma or platelet poor plasma is then automatically drawn out of the centrifuge bag and proportioned into separate chambers in a dispenser. The first portion is activated where a clot is formed and thrombin is obtained. The thrombin is then latter mixed with the second portion to obtain a platelet gel.

The present invention prevents accidental activation of an improper operation that could cause harm to a patient or a shut down of the system by requiring confirmation of each step by an operator. The invention provides screen displays with detailed setup instructions, eliminating the need for secondary documentation that might not be allowed in a surgical environment. The invention also instructs the operator at the appropriate times to do certain manual steps such as opening and closing clamps. Since the opening and closing of clamps is a highly critical operation, confirmation of these steps is also required. A method and suitable apparatus is also disclosed for sequestration of platelet rich plasma whereby a blood sample is spun at a high speed sufficient to separate solid cells from the blood sample and spun at a lower speed for a predetermined time to allow platelets to elute from the solid cells.
