The present invention encompasses a capture membrane comprising a porous filter membrane having a hapten bound directly or indirectly to the membrane wherein complexes formed by specific binding having an anti-hapten bound to a binding member of the specifically binding complex are removed from a solution by the hapten as the solution passes through the membrane. In the preferred embodiment biotin is the hapten and avidin or streptavidin is the anti-hapten.

The method involves contacting a test solution containing the sample, an antibody for the analyte, and a conjugate of the analyte and a label with a contact portion of a strip of bibulous material capable of being traversed by the test solution through capillary action. The strip contains a first receptor capable of binding to said conjugate. The first receptor is non-diffusively bound to a situs on the strip separate from the contact portion of the strip. The strip further contains a second receptor capable of binding the antibody to the analyte between the situs and the contact portion. The second receptor is non-diffusively bound to the strip. At least a portion of the test solution is allowed to traverse the strip by capillary action and thereby contact the situs. The strip is exposed to a signal producing means capable of interacting with the label to produce a signal in relation to the amount of analyte in the test solution. The signal produced at the situs is then detected.

The bibulous material contains a first receptor capable of binding to said conjugate. The first receptor is non-diffusively bound to a situs on the bibulous material separate from the contact portion. The bibulous material further contains a second receptor capable of binding the antibody to the analyte between the situs and the contact portion. The second receptor is non-diffusively bound to the bibulous material. At least a portion of the test solution is allowed to traverse the bibulous material by capillary action and thereby contact the situs. The situs is examined for the presence of the conjugate. To this end, the situs can be exposed to a signal producing means capable of interacting with the label to produce a signal in relation to the amount of analyte in the test solution. The signal produced at the situs is then detected.

This invention is to a method for detecting an amphiphilic antigen in a biological sample suspected of containing the amphiphilic antigen, which method comprises providing in combination a hydrophilic solid support modified to have a hydrophobic surface and an assay medium suspected of containing an amphiphilic antigen, incubating the combination under conditions sufficient for the amphiphilic antigen to bind to the hydrophobic surface, and determining the presence or amount of the amphiphilic antigen bound to the hydrophobic surface.

Background interference which disrupts the measurement of an analyte in enzyme immunoassays with samples is substantially diminished. In accordance with one embodiment of the invention, the serum sample is treated with a sufficient amount of a peracid compound, either organic or inorganic, for a time and under conditions sufficient to reduce or eliminate background interference contributed by serum components other than analyte. The peracid compound is readily quenched, without adversely affecting the assay compositions. In another embodiment of the invention a liqand for the interfering component of the sample, such as unconjugated enzyme which is inactive but antigenic, is mixed with the sample to be analyzed. The presence of the liqand substantially reduces or eliminates background interference from components of the sample other than analyte.
