A method for expressing insecticidal protein structural genes in cotton plant genomes is provided. In the preferred embodiments this invention comprises placing a structural gene for thecrystal protein under control of a plant or a T-DNA promoter and ahead of a polyadenylation site followed by insertion of said promoter/structural gene combination into a plant genome by utilizing anTi plasmid-based transformation system. The modified Ti plasmid is then used to transform recipient plant cells. Also provided are the plants and tissues produced by this method and bacterial strains, plasmids, and vectors useful for execution of this invention.

Insect resistant cotton plants

A method for expressing insecticidal protein structural genes in plant genomes is provided. Insect resistant plants

A nucleic acid sequence encoding for oryzacystatin-I peptides and a signal peptide therefore is provided. The oryzacystatin-I peptide is approximately 12.6 kDa, and is approximately twelve amino acid residues longer than previously described oryzacystatin-I peptides. The nucleic acid sequences may be cloned into vectors, and used to transform plants conferring resistance to plant pests, including insects and nematodes, that utilize cysteine proteases, and to viruses with processing mechanisms involving cysteine proteases.

Oryzacystatin-I applications and methods

The sequence of the T-DNA of the octopine-type Ti plasmid found in Agrobacterium tumefaciens ATCC 15955 is disclosed. Fourteen open reading frames bounded by eukaryotic promoters, ribosome binding sites, and polyadenylation sites were found. The use of promoters and polyadenylation sites from pTi15955 to control expression of foreign structural genes is taught, using as examples the structural genes for the Phaseolus vulgaris storage protein phaseolin, P. vulgaris lectin, thaumatin, and Bacillus thuringiensis crystal protein. Vectors useful for manipulation of sequences of the structural genes and T-DNA are also provided.

The subject invention pertains to materials and methods for transformed plants and plant tissues that are capable of expressing high levels of stable proteins which are localized as protein bodies within the plant cell. Transformed plants co-expressing high levels of both the 15 kD and 10 kD zein proteins are disclosed which accumulate to high levels as protein bodies in the vegetative tissue of the plant. Transformed plants co-expressing the 15 kD and 10 kD zein proteins are useful for providing forage crops containing increased levels of sulfur containing amino acids, such as methionine, in the diet of animals that normally feed on such crops. Also contemplated by the subject invention are transformed plants or plant tissue comprising stable protein bodies which contain heterologous proteinaceous material. In one embodiment, a stable protein body is expressed in a plant or plant tissue as a fusion protein comprising a zein protein and an operably linked protein or peptide. The protein bodies provided in the present invention are resistant to rumin digestion or environmental degradation.

Co-expression of proteins

The invention provides genetically modified plant cells having a plant structural gene introduced and expressed therein under the control of a T-DNA promoter. In particular, the invention provides genetically modified plant cells having the phaseolin gene introduced and expressed therein under the control of a T-DNA promoter. Also provided are novel strains of bacteria containing and replicating T-DNA which has been modified to contain an inserted plant structural gene under the control of a T-DNA promoter. Further, the invention provides novel plasmids having the ability to replicate in E. coli and comprising T-DNA which has been modified to contain an inserted plant structural gene under the control of a T-DNA promoter.

Plant structural gene expression

The phaseolin structural gene and promoter are disposed. Also disclosed are plasmids, bacteria, plant cells, plant tissue and plants containing the phaseolin promoter. The promoter finds use in the expression of heterologous plants genes, including the phaseolin gene, in transformed plant cells and plants.
