Provided is a surface having metal regions and an interstitial region having a composition that differs from the metal regions, wherein a continuous gel layer coats the surface across the metal regions and the interstitial regions. Nucleic acids or other analytes can be attached to the continuous gel layer such that a greater amount is attached over the metal regions than over the interstitial region. Also provided are methods for making such surfaces. Methods are also provided for making an array of nucleic acids or other analytes using such surfaces.

Patterned flow-cells useful for nucleic acid analysis

A method for scanning a microarray, including (a) providing a microarray to an optical scanner, wherein the microarray includes a surface having features having different target molecules; (b) scanning the surface in the x or y direction to acquire optical signals from the features at sequential regions of the surface, wherein the focus setting between the optical scanner and the surface is dynamically controlled in real time by repeatedly (i) adding a predetermined focal offset, thereby introducing an error in the focus setting, and (ii) adjusting the focus setting to reduce the error in the focus setting, thereby acquiring the optical signals from different regions at different degrees of focus such that a subset of the regions that are scanned have an introduced focus error; and (c) analyzing the optical signals that are obtained at the different degrees of focus to identify the different target molecules.

In an example of a method of making a flowcell, an organic solid support including sidewalls and a top is provided. A bottom surface of the organic solid support adjacent to the sidewalls provides a laser bonding foot. In the method, the laser bonding foot is bonded to an inorganic solid support to form a channel having sidewalls and a top defined by the organic solid support.

Where desired, some or all of the computationally intensive tasks may be off-loaded to enhance the portability and speed of the device. A detection apparatus having a read head including a plurality of microfluorometers positioned to simultaneously acquire a plurality of the wide-field images in a common plane; and (b) a translation stage configured to move the read head along a substrate that is in the common plane. The substrate can be a flow cell that is included in a cartridge, the cartridge also including a housing for (i) a sample reservoir; (ii) a fluidic line between the sample reservoir and the flow cell; (iii) several reagent reservoirs in fluid communication with the flow cell, (iv) at least one valve configured to mediate fluid communication between the reservoirs and the flow cell; and (v) at least one pressure source configured to move liquids from the reservoirs to the flow cell. The detection apparatus and cartridge can be used together or independent of each other.

Integrated optoelectronic read head and fluidic cartridge useful for nucleic acid sequencing

A labeled nucleotide includes a nucleotide, a linking molecule attached to a phosphate group of the nucleotide, and a redox-active charge tag attached to the linking molecule. The redox-active charge tag is to be oxidized or reduced by an electrically conductive channel when maintained in proximity of a sensing zone of the electrically conductive channel.

A sensor includes two electrodes and a modulatable electrically conductive channel attached to the two electrodes. The modulatable electrically conductive channel includes a modified, partially double stranded nucleic acid polymer electrically connected to the two electrodes and bridging the space between the two electrodes. The modified, partially double stranded nucleic acid polymer includes two polynucleotide chains partially bonded together, a gap in a first of the polynucleotide chains wherein nucleotide bases are missing, and a plurality of nucleotide bases of a second of the polynucleotide chains exposed at the gap in the first of the polynucleotide chains.
